摘  要:目的:建立气道变态反应性小鼠模型,观察玉屏风散对支气管哮喘小鼠白细胞介素-10(IL-10)和血管细胞黏附分子-1(VCAM-1)表达的影响,探讨玉屏风散对哮喘的治疗机制。方法:40只Balb/c小鼠随机分成对照组、模型组、布地奈德组和玉屏风散组。应用OVA(卵蛋白)诱导炎症模型。RT-PCR法测定肺组织VCAM-1和IL-10的mRNA水平,ELISA法测定肺泡灌洗液(BALF)的IL-10水平。结果:模型组、布地奈德组和玉屏风散组的IL-10 mRNA表达与对照组相比均明显增加,有显著性差异(P<0.05)。玉屏风组,布地奈德组VCAM-1 mRNA的表达与模型组相比明显降低,有显著性差异(P<0.05)。玉屏风散组BALF的IL-10水平与对照组、模型组相比显著升高(P<0.05)。结论:玉屏风干预后,支气管哮喘模型小鼠IL-10水平升高,并显著降低VCAM-1等黏附分子含量,达到治疗哮喘的目的。Objective: To explore the therapeutic mechanism of Yupingfeng San on asthma by observing the effect of this formula on IL-10 and VCAM-1( vascular cell adhesion molecule-1) in mice with bronchial asthma. Methods: 40 Balb/c mice were randomly divided into the blank control group,the model group,Budesonide treatment group and Yupingfeng San treatment group. OVA( ovalbumin) were used to induce the inflammation model. The mRNA levels of VCAM-1 and IL-10 were detected in lung tissue by RT-PCR,and IL-10 level in BALF was measured by ELISA method. Results: Compared to the blank control group,the mRNA expression of IL-10 were significantly increased in the model group,Budesonide group and Yupingfeng San group( P〈0. 05). The expression of VCAM-1 mRNA in Yupingfeng San group and Budesonide group were both significantly lower than that in the model group( P〈0. 05). The IL-10 level in BALF of Yupingfeng San group was significantly higher than that of the blank control group and the model group( P〈0. 05). Conclusion: The increase of IL-10 by intervention with Yupingfeng San is a protective regulation,it can significantly reduce VCAM-1 to treat bronchial asthma.
