摘  要:目的探讨DHA对PC-12细胞的分化促进作用。方法通过CCK8法筛选DHA的最适浓度,免疫荧光法检测神经元标志物MAP2,qPCR检测Beclin、ATG5和LC3基因表达量。Western blotting检测蛋白水平的含量。结果结果显示浓度为0.01、0.1、1μg/mL的DHA干预PC12h,细胞活力和正常组比较无差异。DHA浓度为0.1μg/mL,干预48 h后PC-12细胞内MAP2阳性细胞数显著增多(P=0.029),细胞突触伸长。qPCR结果显示ATG5、Beclin的mRNA表达水平显著升高,LC-3的mRNA表达水平升高,Western blotting结果显示Beclin、ATG5蛋白水平有显著升高。结论低浓度的双氢青蒿素可以通过自噬调节PC12细胞突触伸长,促进细胞分化,对神经退行性等疾病预防和治疗有重要意义。Objective To investigate the effect of DHA on the differentiation of PC-12 cells.Methods The optimal concentration of DHA was screened by CCK8.The neuronal marker MAP2 was detected by immunofluorescence,and the expression levels of Beclin,ATG5,and LC3 were detected by qPCR.Western blotting was used to determine the protein level.Results The results showed that DHA were 0.01,0.1,and 1 mg/mL to induce with PC12,there was no difference with the normal group.After DHA induced in concentrations of 0.1 mg/mL at 48 h,the number of MAP2 positive cells increased significantly(P=0.029) and synaptic of cell elongation in PC-12 cells.qPCR results showed that ATG5,Beclin mRNA expression levels were significantly increased,LC-3 mRNA expression levels increased,Western blotting results showed that Beclin,ATG5 protein levels were significantly increased.Conclusion Dihydroartemisinin can regulate the synaptic elongation of through autophagy,promote cell differentiation in PC12 cells,and play an important role in the prevention of neurodegenerative diseases.
