摘  要:为了从东太平洋深海沉积物中筛选得到具有群体感应抑制活性的细菌菌株,为进一步以抑制病原菌群体感应系统为靶点研究抗感染药物提供新颖、有效的天然产物资源.采用10倍梯度稀释法涂平板从深海沉积物中分离细菌菌株,以紫色杆菌ATCC12472和紫色杆菌CV026报告菌株建立双菌株筛选模型,采用双层平板打孔法筛选具有群体感应抑制活性的菌株.从深海沉积物样本中共分离得到50株细菌,经初筛和复筛,得到了3株具有较强群体感应抑制活性的菌株,具有进一步研究的价值.结果表明:深海细菌中含有具有群体感应抑制活性的菌株,有望成为天然群体感应抑制剂和天然药物先导化合物的重要来源.In order to obtain novel and effective natural product resources for anti-infection drugs targeting the quorum sensing system of pathogens,screening of bacterial strains with quorum sensing inhibitory activity was carried out.The bacterial strains were isolated from deep-sea sediments using a 10-fold dilution coating plate method.A double layer agar diffusion method was established using dual strain Chromobacterium violaceum ATCC12472 and Chromobacterium violaceum CV026 as reporter.Fifty deep-sea bacteria were isolated from the samples,and 3 strains with strong quorum sensing inhibitory activity were obtained.The results showed that many deep-sea bacteria have exhibited the ability of quorum sensing inhibition,which is expected to be an important source of natural quorum sensing inhibitors and natural drug lead compounds.
