摘  要:目的探讨HLA-DR^(-/low)CD33^+CD11b^+CD15^+CD14-多型核粒细胞(PMN,简称CD15^+PMN)在子宫内膜异位症(Endo)中的临床意义。方法用流式细胞术检测正常体检者(HC组,n=7)、不孕症患者(Control组,n=25)及Endo患者(Endo组,n=26)外周血单核细胞(PBMCs)中CD15^+PMN的水平并测定其体外免疫抑制功能,同时检测Control组和Endo组腹水单个核细胞(PFMCs)中CD15^+PMN的水平。结果与HC组和Control组相比,Endo组PBMCs中CD15^+PMN的水平明显升高;与Control组相比,Endo组PFMCs中该细胞群的水平也明显上升,且Endo组患者晚期阶段PBMCs和PFMCs中CD15^+PMN的水平明显高于早期阶段,差异均有统计学意义(P<0.05)。体外T细胞增殖实验发现Endo组患者PBMCs中CD15^+PMN不具有抑制T细胞增殖的能力。结论 Endo患者PBMCs和PFMCs中存在异常增多的CD15^+PMN,提示CD15^+PMN可能参与子宫内膜异位症的发生和发展。Objective To explore the clinical significance of HLA-DR(-/low)CD33+CD11 b+CD15+CD14-polymorphonuclearneutrophils(PMN)in endometriosis(Endo).Methods Seven healthy controls,twenty-five infertile patients and twentysixendometriosis patients were enrolled in this study.The levels of HLA-DR-/lowCD33+CD11 b+CD15+CD14-cells in PBMCs andPFMCs were analyzed by flow cytometry,and the effect of HLA-DR-/lowCD33+CD11 b+CD15+CD14-cells on T cell proliferationwas determined by CFSE dilution.Results The proportion and absolute number of CD15+PMN in PBMCs and PFMCs ofendometriosis patients were significantly increased compared with controls(P〈0.05).There was also significantly increasedin CD15+PMN level of PBMCs and PFMCs at the stage Ⅲ/Ⅳ of endometriosis.Depletion of Endo-derived CD15+PMN fromPBMCs did not influence T-cell response.Conclusion We demonstrated that there exited abnormal expansion of CD15+PMN in PBMCs and PFMCs of patients with endometriosis,suggesting that CD15+PMN may be involved in the developmentof endometriosis.
