摘  要:为研究芍药苷对四氯化碳(CCl_4)诱导的急性肝损伤的保护作用及相关机制。将36只健康雄性C57BL/J小鼠随机分成6组:空白组,芍药苷对照组,CCl_4模型组(0.1%,20 mL/kg),高、中、低剂量芍药苷+CCl_4组(10 mg/kg,30 mg/kg,100 mg/kg),每组6只。24 h后眼球取血收集血清,测定丙氨酸氨基转移酶(ALT),天门冬氨酸氨基转移酶(AST)活性。用苏木精-伊红(HE)染色,观察肝脏的组织学改变;试剂盒测定肝组织中SOD、GSH-PX、CAT的活性及MDA和GSH含量;ELISA法检测血清中TNF-α、IL-6含量;试剂盒检测小鼠肝组织中Caspase-3的活性;q RT-PCR检测肝组织中HO-1 mRNA的表达。实验发现PAE降低小鼠血清ALT、AST水平,改善肝脏的病理形态;芍药苷抑制CCl_4诱导氧化应激,升高肝组织HO-1 mRNA水平;降低TNF-α、IL-6含量;芍药苷减低肝组织中Caspase-3活性,减少肝细胞凋亡。由此可知,芍药苷可保护CCl_4诱导的急性肝细胞损伤,该保护作用可能与抑制脂质过氧化,减少促炎细胞因子产生,减少肝细胞凋亡,促进抗氧化蛋白表达有关。The present study aims to investigate the roles and specific mechanisms of paeoniflorin(PAE) on CCl_4-induced acute liver injury.Mice were randomly divided into six groups(n=6):normal control group,PAE group,CCl_4 group(0.1%,20 m L/kg) and PAE +CCl_4 groups(10 mg/kg,30 mg/kg,or 100 mg/kg).Twenty-four hours after CCl_4 injection,mice were anesthetized and killed,and then serum were collected to detect alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activities.Tissue sections were stained with hematoxylin and eosin(HE) for histopathological evaluation under light microscope;the activities of SOD,GSH-PX and CAT and the content of MDA and GSH in liver tissues were detected using kits following manufacturer's directions;serum of TNF-α and IL-6 levels were detected by ELISA sandwich kits.Activity of caspase-3 in liver tissue was measured using a caspase-3 colorimetric assay kit;q RT-PCR was used to detect the m RNA expression of HO-1 in liver tissue.It was found that paeoniflorin reduced serum ALT and AST levels,pathological examination showed paeoniflorin pretreatment attenuated CCl_4-induced hepatic;Paeoniflorin inhibited CCl_4-induced oxidative stress,alleviated the pro-inflammatory cytokines(TNF-α,IL-6) in CCl_4 exposed mice;Paeoniflorin reduces Caspase-3.activity and decreases hepatocyte apoptosis.All these results suggest that Paeoniflorin could effectively prevent CCl_4-induced acute liver injury by inhibiting lipid peroxidation and pro-inflammatory cytokines production,reducing hepatocyte apoptosis,promoting antioxidant proteins and anti-inflammatory cytokine.
