摘 要:为研究云南产不同色型玛咖对体内能量代谢的影响及机理。将40只SD雌性大鼠按体质量随机分为黑玛咖、紫玛咖、黄玛咖、白玛咖、空白对照5组,其中空白对照组灌胃等体积1%羧甲基纤维素钠溶液,而试验组灌胃0.42g/kg·BW的1%羧甲基纤维素钠玛咖干粉悬浮液,每天1次,连续灌胃28d后,测定血清胰岛素样生长因子(insulin-like growth factor 1,IGF-1)、血糖、肝/肌糖原、血清胰岛素(insulin,Ins)、血清和肝脏组织氨基酸的含量及变化;测定肝脏组织中腺苷酸活化的蛋白激酶(AMP activated protein kinase,AMPK)、哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)、蛋白激酶B(protein kinase B,AKT)和肝激酶B1(liver kinase B1,LKB1)的mRNA水平。试验结果表明:与空白对照组相比,黑玛咖、紫玛咖组试验大鼠相对骨骼肌增重显著(P<0.05),玛咖对各试验大鼠血清Ins和IGF-1含量的影响均不显著;黑玛咖和紫玛咖可显著(P<0.05)上调血清中Asn、Arg、Ser和Leu的含量,显著(P<0.05)上调肝脏中Asp、Ser、Thr、Ala、Val和Leu的含量。各玛咖试验组均显著(P<0.05)上调试验大鼠肝脏mTOR mRNA表达量,黑玛咖和紫玛咖可显著(P<0.05)上调AKT mRNA表达量;各玛咖试验组对AMPK和LKB1mRNA表达量的影响均不显著(P>0.05)。综上可知,玛咖可通过调节能量分配来改变机体的组成,氨基酸可能是介导mTOR活化的主要信号因子。Investigated the effect of different color Maea on energy metabolism of rats. 40 SD female rats were randomly assigned to 5 groups according to their body mass, which includes one control group and four experimental groups. Respectively, the four experimental groups were gavaged with 1 % carhoxymethylcellulose sodium suspension with 0.42 g/kg- BW black Maca, purple Maca, yellow Maca and white Maca powder daily; the control group was gavaged with 1 % carhoxymethylce[lulose sodium solution without Maca pow- der daily. For 28 days, determinated content of blood glucose, glyco gen, insulin and insulin-like growth factor 1 in serum, amino acid content in serum and liver. In addition, detected the mRNA level of AMPK (AMP activated protein kinase), mTOR (mammalian target of rapamycin), AKT (protein kinase B) and LKB1 (liver kinase B1) by qRT PCR (quantitative Real time Polymerase Chain Reaction). These results showed that compared with the control group, relative skeletal muscle mass of rats was significantly increased (P〈0.05), the content of Asn, Arg, Ser and Leu in serum and the content of Asp, Ser, Thr, Ala, Val and Leu in liver also were significantly in creased by black and purple Maca (P〈0.05). However, there is no significant change in the content of Ins and IGF- 1 in serum of rats ga- vaged with black and purple Maca. In the liver of rats, the mRNA level of roTOR were significantly upregulated by four colors Maca (P〈0.05), the mRNA level of AKT were significantly upregulated by black and purple Maca (P 〈 0. 05), however, there is no significant difference in the mRNA levels of AMPK and LKB1 (P〉 0.05). Overall, Maca can change l, ody compositions hy regulating the energy distribution, amino acids may be signal molecules that mediate mTOR activation.
