摘 要:目的:观察针刺预处理对缺血再灌注损伤大鼠心肌HMGB1(高迁移率族蛋白1)表达的影响。方法:40只雄性SD大鼠随机分为4组,假手术组(Sham)、缺血再灌注组(I/R)、电针预处理组(EP)、电针对照组(EC);建立心肌缺血再灌注模型,EP组于造模前1周即开始每天电针大鼠"内关穴",EC组则予电针"外关穴"处理。通过心脏彩超比较造模后各组大鼠心功能,蛋白免疫印迹法检测大鼠心肌HMGB1与STAT3(信号转导和转录活化因子3)的表达。结果:EP组与I/R组相比,左心室前壁厚度、LVDd、LVDs、FS、LVEF均有明显改善(均P<0.01),HMGB1的表达明显减少(P<0.01),p-STAT3的表达显著提高(P<0.01);而EC组与模型组比较,未见明显变化。结论:针刺"内关穴"预处理可抑制心肌组织HMGB1表达发挥心肌缺血再灌注损伤保护作用,其机制可能与STAT3激活相关。Objective:To research the effect of electro-acupuncture preconditioning on the expression of HMGB1( high mobility group protein 1) in rats with myocardial ischemia reperfusion injury.Methods:Forty male SD rats were randomly divided into four groups:Sham operation group( Sham),ischemia reperfusion group( I/R),electro-acupuncture pretreatment group( EP) and electro-acupuncture control group( EC).The model of myocardial ischemia reperfusion was established.EP group received electro-acupuncture at Neiguan point every day one week before the models were established,while EC group was treated with electro-acupuncture at Waiguan point.The cardiac function of rats in each group was examined by Doppler echocardiography,and the expressions of HMGB1 and STAT3( signal transduction and transcription activator 3) in myocardium of rats were detected by Western blot.Results:Compared with I/R group,the thickness of left ventricular anterior wall,LVDd,LVDS,FS and LVEF in EP group were significantly improved( all P〈0.01),the expression of HMGB1 was significantly decreased( P〈0.01),and the expression of p-stat 3 was significantly increased( P〈0.01).There was no significant change between EC group and model group.Conclusion:Electro-acupuncture preconditioning at" Neiguan point"can inhibit HMGB1 expression in myocardial tissue and played a protective role in myocardial ischemia reperfusion injury,the mechanism may be related to STAT3 activation.
