摘 要:目的:通过观察利湿化瘀法对湿瘀型慢性盆腔炎大鼠子宫组织NF-κBp65(核因子蛋白)、IκK(IκB激酶α)蛋白表达的影响,探讨该法的临床作用机理,为临床研究及新药研发提供更深层次的理论依据。方法:用苯酚胶浆注入大鼠两侧子宫造模,成功后,随机分成模型组,中药高、低剂量组和妇乐颗粒对照组。除模型组给予生理盐水灌胃外,其余各组分别给以相应剂量的利湿化瘀中药和妇乐颗粒灌胃,每天1次。21d后,处死大鼠,取材,采用Western Blot法检测大鼠子宫组织NF-κBp65、IκKα蛋白表达。结果:与模型组比较,中药高、低剂量组及妇乐组大鼠子宫组织NF-κBp65表达均减弱,IκKα表达增强,差异有显著性(P<0.05),中药高剂量组效果优于妇乐组,但两组比较,无显著性差异(P>0.05)。结论:利湿化瘀法通过调节大鼠子宫组织NF-κBp65、IκKα蛋白的表达,促进细胞凋亡,减轻盆腔炎症反应。Objective:To explore the mechanism of Removing Dampness and Blood Stasis Method on chronic pelvic inflammatory and provide the deeper theoretical basis for new medicine found by observing the effects on the expressions of NF-κBp65( nuclear factor protein) and IκKα( IκB kinase α) protein in the uterine tissue of rats with chronic pelvic inflammatory disease.Methods:Models of chronic pelvic inflammatory were induced by injecting phenolic glue to rats' bilateral uterus,experiment was divided into normal,model,Fule Granule and Chinese Medicine high and low dose groups,each group was given the corresponding saline,Fule Granule and high,low dose Chinese Medicine once a day for 21 days.Western blotting was applied to determine the NF-κBp65 and IκKα protein expressions in the uterine tissue of rats.Results:The NF-κBp65 protein expressions in the uterine tissue of model group rats was increased significantly and IκKα expression was decreased to compare with normal group( P〈0.05),compared with the model group,the expression of NF-κBp65 in the uterine tissues of the high-dose,low-dose group and the Fule Granules group was weakened,and the expression of IκKα was increased,the differences were significant( P〈0.05).The effect of high-dose group was better than that of the Fule Granules group.But,there was no significant difference between the two groups( P〈0.05).Conclusion:Removing Dampness and Blood Stasis Method can reduce pelvic inflammatory by regulating the expressions of NF-κBp65 and IκKα protein in uterus tissue.
