摘  要:DREB2As是一类可以激活干旱响应基因表达的转录因子。为了提高花生的耐旱性,本研究通过农杆菌介导的转化方式将AlDREB2A转录因子基因导入花生中。同时对影响花生转化的3种因素(烟草提取物添加量、超声波处理时间、抗生素浓度)进行优化。在聚乙二醇的干旱胁迫压力之下,AlDREB2A转基因花生的叶片相对含水量和脯氨酸含量均明显高于非转基因花生,而丙二醛含量低于非转基因花生。在表型方面,转基因花生的萎蔫程度也低于非转基因花生。研究证实,转基因花生比非转基因花生有更高的耐旱性。DREB2 As is a group of transcription factors that activate the expression of drought-responsive genes.To enhance the drought tolerance of peanut plant,AlDREB2 A was transferred into peanut through Agrobacterium transformation in this study.Factors influencing transformation efficiency(dosages of tobacco extracts,ultrasonic treatment time and concentration of antibiotics)were optimized.The results showed that:After drought stress treatment with polyethylene glycol,the relative water content and proline content in transgenic peanuts were notablely higher than those in non-transgenic peanuts,and the content of malondialdehyde was lower than that of non-transgenic peanuts;The wilting degree of transgenic lines was less than that of non-transgenic peanuts.This study demonstrated that the transgenic peanuts had higher drought tolerance ability than non-transgenic peanuts.
