摘 要:目的探讨米非司酮对人早孕绒毛骨桥蛋白(osteopontin,OPN)、绒毛膜促性腺激素-β(humanchorionicghoriogonadotrophin-β,β-HCG)、P53蛋白表达的影响。方法将2016年5月-2017年12月解放军第313医院收治的120例早孕妇女分为A、B、c、D组各30例,分别于人工流产术前服用米非司酮0、100、150、200mg。收集4组术后新鲜绒毛组织,采用免疫组织化学半定量法检测OPN、β-HCG、P53蛋白表达情况。结果B、c、D组的OPN蛋白表达明显低于A组(P〈0.05),C、D组明显低于B组(P〈0.05);A、B、C、D组的β-HCG蛋白表达无明显差异(P〉0.05);B、C、D组P53蛋白表达明显高于A组(P〈0.05),C、D组明显高于B组(P〈0.05)。结论不同剂量的米非司酮对早孕绒毛组织中OPN和P53蛋白表达有显著影响,而对β-HCG蛋白表达无显著影响,推测其抗早孕机制可能与抑制OPN、上调P53的表达有关。Objective To explore the effect of mifepristone on the expression of OPN, β-HCG and P53 protein in chorionic villi of human early pregnancy. Methods 120 cases women with early pregnant from May. 2016 to Dec.2017 were divided into Group A,B ,C and D,30cases in each group,and were separately given mifepristone Omg,10Omg, 150rag and 200rag before artificial abortion. Fresh chorionic tissues after abortion of 4 groups were collected,and the expression of OPN, β-HCG and P53 protein in chorionic villi were detected by immunohistochemistry and semiquantita methods. Results The expression of OPN in Group B ,C and D were all significantly lower than that in Group A (P 〈 0.05),and that in Group C and D were both significantly lower than that in Group B (P 〈 0.05).There were no ignifieantly difference between 4 groups in comparing the expression of β-HCG (P〉 0.05). The expression of P53 in Group B ,C and D were all significantly higher than that in Group A(P〈 0.05), and that in Group C and D were both significantly higher than that in Group B (P 〈 0.05).Conclusions Different doses of mifepristone can significantly effect on the expression of OPN and P53 protein in villi of early pregnancy,while can not significantly effect on the expression of β-HCG. It is speculated that the mechanism of anti early pregnancy may be related to inhibition of OPN and up regulation of P53 expression.
