摘 要:目的探究白藜芦醇对前列腺癌细胞侵袭和凋亡的影响及其分子机制。方法将前列腺癌细胞分为5组,命名为对照组、Z-DEVD-FMK组、Res低剂量组、Res中剂量组、Res高剂量组,采用实时荧光定量PCR(qRT-PCR)检测PC-3细胞中Caspase-3和PARP-1 mRNA表达情况,分别采用Transwell小室和流式细胞仪检测PC-3细胞侵袭和凋亡情况,蛋白免疫印迹(Western blot)检测PC-3细胞中MMP-9表达情况。结果 Res组PC-3细胞中Caspase-3和PARP-1 mRNA表达水平显著高于对照组和Z-DEVD-FMK组(P<0.05),呈剂量依赖性;Res组PC-3细胞侵袭能力显著低于对照组和Z-DEVD-FMK组(P<0.05),呈剂量依赖性;Res组PC-3细胞凋亡率显著高于对照组和Z-DEVD-FMK组(P<0.05),呈剂量依赖性;Res组PC-3细胞中MMP-9表达水平显著低于对照组和Z-DEVD-FMK组(P<0.05),呈剂量依赖性。结论白藜芦醇能够上调Caspase-3和PARP的表达,进而诱导前列腺癌细胞凋亡,为前列腺癌的临床治疗提供一定的理论依据。Objective To investigate the effect of resveratrol on invasion and apoptosis of prostate cancer cells and its molecular mechanism. Methods Prostate cancer cells were divided into five groups,named as control group,ZDEVD-FMK group,Res low-dose group,Res medium-dose group,Res high-dose group; real time quantitative PCR(qRT-PCR) was used to detect the expressions of Caspase-3 and PARP-1 mRNA in PC-3 cells. Transwell cell and flowcytometry were used to detect the invasion and apoptosis of PC-3 cells respectively,and the expression of MMP-9 in PC-3 cells was detected by Western blot. Results The expression levels of Caspase-3 and PARP-1 mRNA of PC-3 cells in Res group were significantly higher than those in control group and Z-DEVD-FMK group(P〈 0. 05),in a dose-dependent manner; the invasive ability of PC-3 cells in Res group was significantly lower than that in control group and Z-DEVD-FMK group(P 〈0. 05),in a dose-dependent manner; the apoptosis rate of PC-3 cells in Res group was significantly higher than that in control group and Z-DEVD-FMK group(P〈 0. 05),in a dose-dependent manner;the expression level of Res in PC-3 group was significantly lower than that in control group and Z-DEVD-FMK group(P 〈0. 05),in a dose-dependent manner. Conclusion Resveratrol can up-regulate the expressions of Caspase-3 and PARP,and induce apoptosis of prostate cancer cells,which provides a theoretical basis for the clinical treatment of prostate cancer.
