摘 要:目的探讨钙调神经磷酸酶(CaN)-活化T细胞核因子(NFATc)信号通路在5-氮杂胞苷(5-aza)诱导人脐带间充质干细胞(hUCMSCs)分化为心肌细胞过程中的作用。方法收集足月顺产健康产妇的脐带,于无菌条件下剪碎后体外分离、培养hUCMSCs,采用10μmol/L或20μmol/L5-aza对P3代hUCMSCs诱导24h后继续培养4周。采用RT-PCR和WesternBlot检测诱导前、后CalcineurinA、GATA4和NFATc2表达。结果与诱导前和对照组比较,10μmol/L和20μmol/L浓度诱导4周hUCMSCs的CalcineurinA、GATA4和NFATc2表达均明显上调(P<0.05或P<0.01)。10μmol/L浓度的5-aza诱导效果效果明显优于20μmol/L(P<0.05或P<0.01)。对照组4周后CalcineurinA、GATA4和NFATc2表达水平较诱导前无明显变化(P>0.05)。结论 CaN-NFATc信号通路参与hUCMSCs心肌细胞分化的调控,10μmol/L的浓度是5-aza诱导hUCMSCs心肌细胞分化的最佳浓度。Objective To investigate the role of calcineurin(CaN)-nuclear factor of activated T cells(NFATc) signaling pathway in the differentiation of human umbilical cord derived mesenchymal stem cells(hUCMSCs) into cardiomyodytes induced by 5-azacytidine(5-aza). Methods The umbilical cord of full-term health maternal were collected, hUCMSCs were isolated and cultured in vitro after sterilizing. The 10 μmol/L or 20 μmol/L 5-aza was used to induce P3 hUCMSCs for24 h and continued for 4 weeks. RT-PCR and Western Blot were used to detect the expression of Calcineurin A, GATA4 and NFATc2 before and after induction. Results After 4 weeks, the expression levels of Calcineurin A, GATA4 and NFATc2 in hUCMSCs induced by 10 μmol/L and 20 μmol/L 5-aza significantly increased than those before induction and the control group(P0.05 or P0.01); After 4 weeks, the effect of 5-aza induction at 10 μmol/L was significantly better than that at 20 μmol/L(P 0.05 or P 0.01). The expression levels of Calcineurin A, GATA4 and NFATc2 in the control group were not significantly different from those before induction(P 0.05). Conclusion CaN-NFATc signaling pathway maybe participate in regulating the differentiation of hUCMSCs into cardiomyodytes, the concentration of 10 μmol/L is the optimal concentration of 5-aza induced differentiation of hUCMSCs into cardiomyodytes.
