摘 要:目的:观察益智仁炮制前后分别组成缩泉丸对大鼠下丘脑-垂体-甲状腺轴(HPT轴)功能的影响,拟阐明缩泉丸中益智仁盐炙前后的科学内涵。方法:取SD大鼠随机分成4组。采用腺嘌呤灌胃复制肾阳虚多尿模型,造模成功后开始分别给予益智仁盐炙前后组成的缩泉丸,空白组和模型组给予等量蒸馏水。实验结束后股动脉取血同时取下丘脑,测定各组大鼠血清三碘甲酸原氨酸(T3)、甲状腺素(T4)、促甲状腺激素(TSH)、血清和下丘脑促甲状腺激素释放激素(TRH)。结果:与空白组比较,模型组大鼠血清中T3、T4、TSH、TRH以及下丘脑TRH含量均显著降低(P<0.05)。与模型组比较,缩泉丸益智仁生品组以及缩泉丸益智仁盐炙品组血清T3、T4、TSH、TRH以及下丘脑TRH含量均显著升高(P<0.05),且盐炙品组含量高于生品组含量(P<0.05)。结论:腺嘌呤所致的肾阳虚模型存在下丘脑-垂体-甲状腺轴功能紊乱,缩泉丸中益智仁盐炙前后对肾阳虚模型的甲状腺功能均有不同程度的改善作用,尤以缩泉丸中益智仁盐炙品效果较好,说明缩泉丸中益智仁用盐炙品的合理性。Objective: To observe the effect of stir-frying with salt-water of Yizhiren in Suoquan Wan on the function of hypothalamus pituitary thyroid(HPT) axis in rats and clarify the scientific connotation Yizhiren in Suoquan Wan by stir-frying with salt-water. Method: SD rats were randomly divided into 4 groups: blank group, model group, Suoquan Wan crude group and Suoquan Wan salt-processed group. Kidney-yang deficiency polyuria rats model was induced by adenine. Then Suoquan Wan crude group and Suoquan Wan salt-processed group were given Suoquan Wan crude and processed product. The blank group and model group were given equal volume of distilled water. Finally, the rat femoral artery blood and the hypothalamus was collected and the contents of rat serum triiodothyronine orthoformate ammonia acid(T3), thyroxine(T4), promoting thyroid stimulating hormone(TSH), serum and hypothalamus promote thyroid hormone releasing hormone(TRH) were detected. Result: Compared with the blank group, the contents of serum T3, T4, TSH, TRH and hypothalamic TRH in the model group were significantly decreased(P〈0.05). Compared with the model group, the levels of serum T3, T4, TSH, TRH and hypothalamic TRH in Suoquan Wan crude group and Suoquan Wan salt-processed group were significantly increased(P〈0.05) and salt-processed product group showed better effect than the Suoquan Wan crude group(P〈0.05). Conclusion: The dysfunction of HPT axis is observed in adenineinduced kidney-yang deficiency polyuria rats model. Suoquan Wan crude group and Suoquan Wan salt-processed group model both improve the thyroid function of kidney-yang deficiency rat, and the salt-processed Yizhiren has better effect. The results indicate the rationality of stir-frying with salt-water of Yizhiren in Suoquan Wan.
