IL-1B和IL-6启动子区基因多态性与宁夏地区回、汉族人群慢性牙周炎的相关性研究

摘　　要：目的:探讨白细胞介素-1B(IL-1B)和白细胞介素-6(IL-6)基因启动子区的基因多态性与宁夏地区回、汉族人群慢性牙周炎(CP)之间的关系。方法:选取宁夏地区CP患者回族132例、汉族141例和健康对照者回族138例、汉族146例,分别抽取外周静脉血;用PCR-RFLP方法,检测各受试者IL-1B-511及IL-6-572两个基因启动子区SNPs多态性位点的基因型,计算其基因型频率和等位基因频率。分析IL-1B基因及IL-6基因启动子区多态性位点的基因型和等位基因频率在回、汉族中的分布差异。结果:(1)回、汉族病例组与健康对照组在IL-1B-511基因型和等位基因频率的分布均有显著性差异(P<0.05);而在IL-6-572基因型和等位基因频率的分布均无显著性差异(P>0.05);(2)回、汉族病例组在IL-1B-511、IL-6-572的基因型和等位基因频率的分布组间均无显著性差异(P>0.05)。结论:IL-1B-511(rs16944)位点的多态性与宁夏地区回、汉族人群CP的易感性具有相关性,在回、汉两民族CP患者之间无统计学差异。IL-6-572(rs1800796)位点多态性与宁夏地区回、汉族人群CP的易感性之间无相关性。AIM： To determine the association between gene polymorphisms in the promoter of interleukin - 1B （IL - 1B） and Interleukin - 6 （IL - 6） and chronic periodontitis（CP） in Hui and Han populations from Ningxia. METHODS： Peripheral venous blood was collected from 132 patients with CP and 138 healthy controls （HCs） of Hui nationality; 141 patients with CP and 146 HCs of Han nationality. The genotypes of IL - 1B-511 （rs16944） and IL - 6 - 572 （rs1800796） were examined by PCR-RFLP respectively. The genotypes and alleles frequency of IL - 1B - 511 and IL - 6 - 572 from all the subjects and the association of gene polymorphisms with CP were analyzed by case-control method, the distributions of genotypes and allele frequencies of IL - 1B gene promoter polymorphism and IL-6 gene promoter polymorphism between the two nationalities was analyzed. RESULTS： There was statistically significant difference of the frequencie of IL - 1B - 511 genotype and allele between the CP patients and HCs in Hui and Han nationalities （ P 〈0.05）. There was no statistically significant difference of the frequencie of IL - 6 - 572 genotype and allele between the CP patients and HCs in both no statistic significance in the frequencies of IL - 1B - 511 and IL - 6 - 572 genotypes and alleles between the Hui CP and Han CP groups. CONCLUSION： The polymorphism of the IL-1B-511 is associated with CP in Hui and Han nationalities, but there was no significant difference between Hui and Han ethnicity. The polymorphism of the IL - 6 - 572 is not associated with CP in both nationalities.
