摘 要:目的:研究不同剂量钴-60(60Co)射线辐照对白术药材4种有效成分含量的影响。方法:采用HPLC-DAD梯度洗脱-波长切换法;以Agilent C18柱(250 mm×4.6 mm,5μm)为色谱柱,流动相为乙腈-水,梯度洗脱,流速为1.0 ml·min-1,柱温35℃,检测波长为220 nm(白术内酯Ⅱ、白术内酯Ⅲ及苍术酮)和275 nm(白术内酯Ⅰ),进样量为10μl。分别选择剂量为5,8,10 k Gy对白术药材进行辐照,比较辐照前后4种有效成分含量的变化。结果:白术内酯Ⅰ、白术内酯Ⅱ、白术内酯Ⅲ、苍术酮分别在0.011~0.217μg,0.011~0.212μg,0.012~0.243μg,0.094~1.885μg范围内有良好线性,平均加样回收率分别为101.3%,100.6%,100.7%和100.5%。经过5,8,10 k Gy辐照后,白术药材所含的白术内酯Ⅰ、Ⅱ、Ⅲ及苍术酮均有变化。超过5 k Gy后,苍术酮辐照前后含量变化差异有统计学意义(P<0.05)。结论:所建立的白术有效成分测定方法回收率高,重复性好,简单实用,可用于白术的质量控制。在60Co辐照剂量不超过5k Gy时,各成分变化不显著,可为白术药材辐照灭菌手段提供参考。Objective: To investigate the effects of different doses of Co-60 irradiation on four kinds of effective components in atractylodis macrocephalae rhizoma. Methods: An HPLC-DAD gradient elution-wavelength switching method was adopted. An Agilent C18 column( 250 mm × 4. 6 mm,5 μm) was used and the mobile phase was acetonitrile-water with gradient elution at the flow rate of1. 0 ml·min-1. The detection wavelength was 220 nm for atractylodesⅡ,atractylodes Ⅲ and atractylone,and 275 nm for atractylodesⅠ,and the column temperature was 35℃. Different doses of 5,8 and 10 k Gy were selected to compare the content changes of the four active components before and after the irradiation,and the difference was investigated by paired t-test. Results: The linear range of atractylodesⅠ,atractylodes Ⅱ,atractylodes Ⅲ and atractylone was 0. 011-0. 217 μg,0. 011-0. 212 μg,0. 012-0. 243 μg and 0. 094-1. 885 μg,respectively,and the average recovery was 101. 3%,100. 6%,100. 7% and 100. 5%,respectively. The contents of atractylodesⅠ,atractylodesⅡ,atractylodes Ⅲ and atractylone were all changed after the irradiation. When the dose of irradiation was more than 5 k Gy,the difference in the content change of atractylone before and after the irradiation had statistical significance( P〈0. 05). Conclusion: The method is with high recovery,good repeatability and simple practicality,which can be used for the quality control of atractylodis macrocephalae rhizoma. When the irradiation dose is not more than 5 k Gy,no significant changes are found in the contents of the components,which provide reference for the irradiation sterilization for atractylodis macrocephalae rhizoma.
