摘  要:目的:建立一种可同时测定复方天麻颗粒中天麻素和五味子醇甲含量的HPLC法。方法:色谱柱为Agilent Zorbax SB-C18柱(150 mm×4.6 mm,5μm),以乙腈和0.1%磷酸溶液为流动相,进行梯度洗脱,流速为1.0 ml·min-1,检测波长为220nm,柱温为30℃,进样量为10μl。结果:天麻素和五味子醇甲分别在8.00~48.00μg·ml-1和4.75~28.50μg·ml-1范围内呈现良好的线性关系,r分别为0.999 9和0.999 5;平均加样回收率分别为99.2%和98.8%。结论:本研究建立了能同时测定复方天麻颗粒中天麻素和五味子醇甲含量的高效液相色谱法,该方法操作简便、稳定可靠,为复方天麻颗粒的内在质量控制提供了新的选择。Objective: To develop an HPLC method for the simultaneous determination of gastrodin and schisandrin in compound Tianma granules. Methods: An Agilent Zorbax SB-C18( 150 mm × 4. 6 mm,5 μm) chromatographic column was used,the mobile phase consisted of acetonitrile and 0. 1% phosphoric acid solution with gradient elution at a flow rate of 1. 0 ml·min-1,the detection wavelength was 220 nm,the column temperature was set at 30℃ and the injection volume was 10 μl. Results: The linear range of gastrodin and schisandrin was 8. 0-48. 0 μg·ml-1( r = 0. 999 9) and 4. 75-28. 50 μg·ml-1( r = 0. 999 5),respectively. The average recovery of gastrodin and schisandrin was 99. 2% and 98. 8%,respectively. Conclusion: An HPLC method for the simultaneous determination of gastrodin and schisandrin in compound Tianma granules was established. The method is simple,stable and reliable,and provides a new choice for the internal quality control of compound Tianma granules.
