摘 要:目的探讨在慢性全脑缺血的状态下,不同浓度的黄芩茎叶黄酮(SSTF)提取物对VD大鼠学习记忆障碍3Zp—tau、PP2AXcGSK313蛋白表达的影响。方法BCCAO建立VD模型,WB法检测p—tau、PP2A及GSK3β蛋白的表达。结果与正常大鼠比较,VD大鼠逃避潜伏期时间延长,穿越平台的次数明显减少,游行距离的百分比明显缩短(P〈0.01),提示VD大鼠存在认知功能障碍。给予SSTF100mg/kg干预治疗后,能够明显改善VD大鼠的认知功能障碍。100mg/kgSSTF能够显著降低VDA鼠磷酸化tau蛋白的水平。进一步研究发现SSTF不能改变VD大鼠总GSK3β蛋白的水平,却能显著提高p—GSK3β蛋白的水平。另外,100mg/kgssTF能够显著改善VD大鼠脑PP2A—subB蛋白表达的水平。结论ssTF是通过对p—tau、PP2A及GSK3B等蛋白的调节而起到预防血管性痴呆的作用。Objective In this study, we want to investigate the effects of scutellaria baicalensis stem-leaf flavonoids ( SSTF ) on the expression of p-tau, PP2A and GSK313 protein in chronic cerebral ischemia-induced vascular dementia rats. Methods BCCAO was used to establish vascular dementia model and WB method was used to detect the expressions ofp-tau, PP2A and GSK3 protein. Results We found that, during the training trail in the Morris water maze assessment, the latency time in the disease model significantly extended after ischemia compared with sham and control- operated animals ( P〈0.05 ) . The percentage of searching time in the sham group was notably higher compared with the VaD group ( P〈0.05 ) . Searching distance in target quadrant, significantly decreased in VaD animals (P〈0.01) . The significant increase of phosphorylated tau was observed in both the cortex and hippocampus after a global ischemia in VaD rats, the phosphorylation of tau was significantly decreased with SSTF treatment similarly in both the cortex and hippocampus. Our resuks showed phosphorylated GSK3 β ( p-GSK3β ) , was significantly down-regnlated in VaD rats by the treatment with 100 mg/kg of SSTF. Similarly, no difference of Cdk5 levels was observed in both the cortex and hippocampus among all groups. However, SSTF administration decreased the levels of p35 and p25, which may ameliorate the activity of Cdk5. PP2A-sub B expression in the cortex and hippocampus was significantly down-regulated in VaD rats. Notably, SSTF reversed the reduction of PP2A-sub B, suggesting that tau phosphorylation might be reduced by SSTF through increasing the PP2A regulation. Conclusion The scutellaria baicalensis stem-leaf flavonoids can play the important role in preventing vascular dementia by regulating the p-tau, PP2A and GSK3 β protein.
