摘 要:目的探讨大蓟醇提物对败血症休克大鼠的影响及其可能的作用机制。方法实验按wichterman方法复制败血症休克模型,随机分为假手术组、模型组、大蓟低剂量组、大蓟高剂量组,用BL-420F+生物机能实验系统检测大鼠心功能;酶联免疫吸附试验(ELISA)法检测血清中TNF-α、IL-1β和IL-6的含量。结果与假手术组比较,模型组大鼠出现低动力学表现,其平均颈动脉压、左心室收缩压、左心室升高最大变化速率和左心室降低最大变化速率均下降,左心室舒张末压升高(P<0.05);与模型组比较,大蓟组大鼠心功能下降,大蓟低剂量组平均颈动脉压、左心室收缩压、左心室升高最大变化速率和左心室降低最大变化速率均下降,左心室舒张末压升高,差异无统计学意义(P>0.05);大蓟高剂量组平均颈动脉压、左心室收缩压、左心室升高最大变化速率和左心室降低最大变化速率均下降(P<0.05),左心室舒张末压升高,差异有统计学意义(P<0.05)。与假手术组相比,模型组血清中TNF-α、IL-1β和IL-6的含量升高,差异有统计学意义(P<0.05);与模型组相比,大蓟低剂量组和大蓟高剂量组血清中TNF-α、IL-1β和IL-6的含量没有明显改变(P>0.05)。结论大蓟对大鼠败血症休克表现出明显的降压作用,可抑制心率和心脏收缩能力,降低大鼠的心功能。在此过程中大蓟的抗菌抑菌作用表现并不明显,未能有效控制炎症感染及炎性因子释放。Objective To investiyate the effects of Daji alcohol extract on septic shock rats and its possible mechanism.Methods Used the wichterman method to make the sepsis shock model,the rats were randomly divided into four groups,including the sham operation group,the sepsis shockgroup,the low dose group of Da Ji and the high dose group of Da Ji. The BL-420 F + was used to test the heart function of rat and the enzyme linked immunosorbent assay( ELISA) method was used to detect the content of TNF-alpha,IL-1 and IL-6 in serum. Results Compared with the sham operation group,the sepsis shock group showed low dynamic performance. The average arterial pressure,the left ventricular systolic,the left ventricle increasing maximum rate of change and the left ventricle decreasing maximum rate of change all decreased,the left ventricular dilation elevated( P〈0. 05). Compare to the sepsis shock group,the cardiac function of rats in the treatment group decreased. The mean arterial pressure,the left ventricular systolic pressure,the left ventricle increasing maximum rate of change and the left ventricle decreasing maximum rate of change decreased in the low dose group,the left ventricular diastolic pressure increased,the differences were not significant( P〈0. 05). Compared with the sham operation group,the content of TNF-alpha,IL-1 beta and IL-6 increased in the serum of the sepsis shock group. The difference was significant( P〈0. 05). Compared with the sepsis shock group,the levels of TNF-α、IL-1β and IL-6 were not significantly altered in the different dose group of Da Ji. Conclusion The Da Ji has obvious depressurization on septic shock rats. It can inhibite the heart rate and the heart contraction,decrease the cardiac function in rats. In the process,the antibacterial activity of Da Ji is not obvious,and it can not control inflammatory infection and the release of inflammatory factor.
