摘 要:目的探讨布拉氏酵母菌对人轮状病毒(HRV)腹泻乳鼠诱导型一氧化氮合酶(i NOS)mRNA表达的影响。方法 ICR乳鼠54只,42只构建腹泻模型(HRV Wa株悬液),另外12只为阴性对照组(等量0.9%Na Cl)。按照体重将建模成功乳鼠分为3组,每组14只。模型组、实验组及阳性对照组分别予以0.9%Na Cl 100μL、布拉氏酵母菌散10^(10)CFU(100μL)和0.1 g·m L^(-1)病毒唑混悬液50μL。阴性对照组予以0.9%Na Cl 100μL。各组均连续灌胃3 d。观察各组腹泻、死亡及粪便中HRV阳性持续时间;用免疫组化法检测小肠组织中干扰素γ(IFN-γ)和细胞间黏附分子-1(ICAM-1)的表达;用实时荧光定量聚合酶链式反应检测小肠组织中i NOS mRNA表达量。结果实验组IFN-γ和ICAM-1阳性细胞百分比分别为(49.51±5.22)%,(43.22±4.51)%,光密度值分别为135.16±3.54,101.64±2.13。模型组IFN-γ和ICAM-1阳性细胞百分比及光密度值高于阴性对照组(P<0.01),实验组和阳性对照组IFN-γ阳性细胞百分比及光密度值高于模型组,ICAM-1阳性细胞百分比及光密度值低于模型组(P<0.05或P<0.01)。阴性对照组、模型组、实验组和阳性对照组小肠组织中i NOS mRNA相对表达量分别为1.00±0.00、1.69±0.22、1.42±0.18、1.37±0.12,模型组小肠组织中i NOS mRNA相对表达量高于阴性对照组(P<0.01),实验组和阳性对照组均低于模型组(均P<0.01)。结论布拉氏酵母菌具有较好的免疫保护作用,可有效降低HRV感染乳鼠腹泻率及死亡率,其机制可能与提高小肠组织中IFN-γ蛋白表达量,降低ICAM-1蛋白和i NOS mRNA表达量有关。Objective To investigate the effect of saccharomyces boulardii on the expression of inducible nitric oxide synthase( i NOS) mRNA in neonatal mice with human rotavirus( HRV). Methods Fifty-four SPF grade ICR neonatal mice were enrolled into the study. Forty-two mice were used to establish diarrhea model( treated with HRV Wa strain suspension) while the other 12 mice were include in the negative control group( treated with equal amount of normal saline). The neonatal mice successfully modeled were divided into 3 groups according to body weight( n = 14) as follows: model group( 100 μL saline),test group [200 μL of saccharomyces boulardii( 10~10 CFU) ],positive control group [50 μL of ribavirin suspension( 0. 1 g · m L^(-1)) ]. The negative control groupwas given 100 μL of normal saline. All groups received oral administration for 3 days. Diarrhea,death and fecal HRV positive duration of each group were observed,immunohistochemistry was used to detect the expressions of γ-interferon( IFN-γ) and leukocyte adhesion molecule-1( ICAM-1) in small intestine of neonatal mice,Real-time quantitative PCR was used to detect the expression of inducible nitric oxide synthase( i NOS) mRNA in small intestine of mice. Results The percentages of IFN-γ and ICAM-1 positive cells in test group were( 49. 51 ± 5. 22) %,( 43. 22 ± 4. 51) %,and the optical density values were 135. 16 ± 3. 54 and 101. 64 ± 2. 13 respectively. The percentage of IFN-γ and ICAM-1 positive cells and the gray values in model group were higher than those in negative control group( P 0. 01),the percentages and the gray values of IFN-γ positive cells in test group and positive control group were higher than those in model group,ICAM-1 positive cells and gray value were lower than those in model group( P 0. 05 or P 0. 01).
