摘  要:目的:探究淫羊藿苷对老龄大鼠骨质疏松骨折的治疗作用和机制。方法:老年雌性大鼠60只,分为空白组,正常组,模型组,低、中、高剂量(40、80、120mg/kg)淫羊藿苷治疗组(低、中、高剂量组)。除空白组外各组均摘除卵巢,3个月后除正常组外,其余各组均建立股骨骨折模型。随后每日治疗组给予相应剂量的淫羊藿苷治疗12周。末次给药后测量血清中骨转化指标,检测大鼠股骨的骨密度,并对股骨进行病理学检查。Western blot法和免疫组化法检测骨组织中Notch1、OPG、RANK和RANKL表达水平。结果:高剂量组的骨小梁排列紧密,骨密度显著回升,血清中的碱性磷酸酶与抗酒石酸性磷酸酶的含量显著降低(P<0.01),骨钙素、雌二醇和骨形成蛋白-2含量明显提升(P<0.01)。另外,淫羊藿苷具有提高Notch1和OPG,抑制RANK和RANKL表达的作用。结论:淫羊藿苷对老龄大鼠骨质疏松骨折具有良好的治疗作用,其主要作用机制与调节Notch1信号通路和OPG/RANK/RANKL系统有关。Objective: To investigate the treatment effect and mechanism of icariin (ICA) on osteoporotic fracture in senile rats. Methods: A total of 60 old female rats were divided into 6 groups: blank group, normal control (NC) group, model control group (MC), treated groups for low/middle/high dosages (ICA-L/M/H). Ovaries were removed from all groups except the blank group. Three months later, osteoporotic fracture model was made in every group except for NC group. And then the ICA-L/M/H groups were received 40, 80 and 120mg/kg icariin every day for 12 weeks. After the last administration, the bone transformation index in serum was measured, the bone density of femur in rats was measured, and the femur was pathologically examined. Western blot and immunohistochemistry analysis were used to detect the expression of Notchl and OPG/RANK/ RANKL system in bone. Results: The trabecular bone of high dose treatment group was closely packed, and the bone density was significantly rebounded, and the contents of serum alkaline phosphatase and tartaric acid resistance phosphatase were significantly decreased, but the contents of osteocalcin, estradiol and bone morphogenetic protein-2 were improved significantly. In addition, ICA could significantly down-regulate the levels of RANK and RANKL and up-regulate the contents of Notchl and OPG. Conclusion: ICA has a good therapeutic effect on osteoporotic fracture in aged rats, and its main mechanism is related to regulating Notch 1 signalling pathway and 0 system.
