摘 要:目的:探讨麝针疗法对缺血性脑中风大鼠卒中后运动神经功能的康复作用及机制。方法:采用改良线栓法建立缺血性脑中风大鼠模型,造模成功大鼠随机分为模型组和土家族麝针组,同时设立假手术组。土家族麝针组以自制麝针刺激大鼠对侧头皮运动区肌层,持续治疗3个疗程;假手术组和模型组不做任何处理。治疗结束后,取各组大鼠评定运动功能,采用ELISA法检测缺血灶周围脑组织内的cAMP和PKA表达,免疫组织化学法检测脑组织中Fos和pCREB蛋白的分布,Western Blot法检测脑组织中pCREB蛋白的表达。结果:与假手术组比较,模型大鼠的运动功能显著降低,其脑组织中cAMP、PKA和Fos的表达显著增强,pCREB的表达显著减弱;与模型组比较,土家族麝针疗法能显著促进缺血性脑中风大鼠的运动功能的康复和神经活性增强标志物Fos蛋白的表达,增强脑组织中cAMP和pCREB、PKA的合成和释放。结论:脑缺血后神经功能具有一定的自然恢复能力,其可能与脑组织中cAMP、PKA的表达增强有关;土家族麝针疗法可能是通过激活Camp/PKA-pCREB信号通路促进缺血性脑中风大鼠运动功能康复。Objective: To explore the rehabilitation effect and mechanism of Tujia musk acupuncture on motor function after stroke in rats with ischemic stroke. Methods: The rat model of ischemic stroke was established by reforming longa method. Then they were randomly divided into model group and tujia musk acupuncture group, and sham operation group was established at the same time. Tujia musk acupuncture group stimulated the rats' muscle layer of the contralateral scalp movement area with self-made musk acupuncture, and continued treatment for 3 courses. The sham operation group and the model group did not do any treatment. After the treatment, the rats in each group were evaluated for motor function, and the expression of cAMP and PKA in the brain tissue around the ischemic lesion were detected by ELISA. The distribution of Fos and pCREB protein in the brain tissue was detected by immunohistochemistry. The expression of pCREB protein was detected by Western Blot method. Results: Compared with the sham operation group, the motor function of the model rats was significantly decreased, and the expression of cAMP, PKA and Fos in the brain tissue was significantly enhanced, and the expression of pCREB was significantly attenuated; Tujia acupuncture therapy can significantly promote the recovery of motor function and the expression of Fos protein, a marker of enhanced neuronal activity in ischemic stroke rats, and increase the synthesis and release of cAMP, pCREB and PKA in brain tissue. Conclusion: Post-ischemic neurological function has a certain natural resilience, which may be related to the increased expression of cAMP and PKA in brain tissue; Tujia musk acupuncture may promote rehabilitation of motor function of ischemic stroke rats by activating Camp/PKA-pCREB signaling pathway.
