当归补血汤有效组分配伍对肝星状细胞Ⅲ型胶原分泌及转化生长因子β1受体的影响

摘　　要：目的观察当归补血汤有效组分及其配伍对大鼠肝星状细胞HSC-T6Ⅲ型胶原分泌和转化生长因子β1(TGF-β1)Ⅰ型和Ⅱ型受体表达的影响。方法当归的有效成分阿魏酸和黄芪的有效成分黄芪甲苷单独及联合处理HSC-T6细胞24 h,ELISA检测培养上清液中Ⅲ型胶原含量,Western blot检测HSC-T6细胞Ⅰ和Ⅱ型TGF-β受体蛋白表达,实时定量PCR检测HSC-T6细胞Ⅰ型TGF-β受体mRNA表达。结果阿魏酸显著抑制了HSC-T6细胞培养上清液中Ⅲ型胶原含量,下调了Ⅰ和Ⅱ型TGF-β受体蛋白表达和Ⅰ型TGF-β受体mRNA表达(均P<0.05),黄芪甲苷对这些指标无显著性影响(均P>0.05)。与阿魏酸单独处理组的作用效果比较,联合处理组无显著性提高(均P>0.05)。结论当归补血汤的有效组分阿魏酸抑制了肝星状细胞Ⅲ型胶原分泌及TGF-β受体表达。Objective The present study attempted to investigate the effect of active component compatibility of DangguiB uxue Decoction on secretion of type Ⅲ collagen and gene expression of transforming growth factor-β1（TGF-β1）type Ⅰ and type Ⅱ receptors in hepatic stellate cell-T6.Methods Hepatic stellate cell（HSC）-T6 cells were treated with astragaloside Ⅳ andferulic acid,either alone or in combination,for 24 h,and then cells were collected.Type Ⅲ collagen secreted by the stellate cells into the culture media was determined by ELISA.Protein expression of TGF-βtype Ⅰ receptor and type Ⅱ receptor were analyzed by Western blot.Messenger RNA expression of TGF-βtype Ⅰ receptor was analyzed by real-time PCR.Results Ferulic acid but not astragaloside Ⅳ inhibited type Ⅲ collagen secreted by the stellate cells into the culture media,down-regulated the protein expression of TGF-βtype Ⅰ receptor and type Ⅱ receptor,and mRNA expression of TGF-βtype Ⅰ receptor.The combination treatment of ferulic acid and astragaloside Ⅳ fail to cause a greater inhibitory effect than ferulic acidalone.Conclusion Ferulic acid,an active component of Danggui Buxue Decoction,suppresses collagen Ⅲ secretion and TGF-βreceptors expression in HSC-T6.
