摘  要:目的观察三七注射液对阿霉素诱导的肾纤维化大鼠血清补体C3a、C5a和肾组织Toll样受体(TLR)1、TLR2的影响。方法将144只SD大鼠随机分为4组:正常组(n=36)、假手术组(n=36)、模型组(n=36)和三七注射液组(n=36),每组在给药后第4周、8周、12周三个时间点各处死大鼠12只。Western blot检测肾组织中TLR1、TLR2蛋白的表达,ELISA法检测血清C3a、C5a的含量。结果与假手术组对比,模型组大鼠BUN和Scr、血清C3a和C5a、肾组织TLR1、TLR2蛋白表达的水平在各时间点均升高,差异有统计学意义(P<0.05或P<0.01)。与模型组组对比,三七注射液组大鼠BUN和Scr、血清C3a和C5a、肾组织TLR1、TLR2蛋白表达的水平在各时间点均下降,差异均有统计学意义(P<0.05或P<0.01)。结论三七注射液可能通过下调TLR1、TLR2蛋白表达的水平,同时降低C3a、C5a的水平,调节补体–炎症受体系统,从而发挥抗肾纤维化作用。Objective To observe the effect of Sanqi injection on the Toll-like receptor TLR1,TLR2 and the serum level of complement system C3 a,C5 a in rats model with Adriamycin-induced kidney fibrosis. Methods 144 rats were divided into 4 groups randomly: normal group,sham group,model group and Sanqi injection group. All the 12 rats in each group were Sacrificed at the time point of 4,8,12 weeks after the treatment. The protein expression of TLR1 and TLR2 were detected through western blot. And the serum level of C3a,C5a was detected by ELISA. Results The level of BUN,Scr,C3a,C5a,the protein expression of TLR1,TLR2 in the kidney in the Model group are increased,there is statistical significance when compared with those in the sham group. However,the level of BUN,Scr,C3a,C5a,the protein expression of TLR1,TLR2 in the kidney in the Model group are decreased,there is statistical significance when compared with those in the model group. Conclusion Sanqi injection could provide a nephroprotective role through down-regulating the protein expression of TLR1,TLR2,and decreasing the serum of C3a,C5a,suggesting that it provides a regulatation between the Toll-like receptor and the complement system.
