摘 要:目的采用斑马鱼模型比较白芍不同部位、主要成分及主要代谢产物的促血管和抗血栓生成作用。方法采用Tg(VEGFR2:GFP)系荧光转基因斑马鱼为模型动物,分别将白芍总样、不同萃取部位、芍药苷、白芍苷和芍药苷代谢素-I设置三个药物剂量组,用荧光显微镜观察并计数不同给药组有血流的斑马鱼体节间血管数;以三氯化铁为血栓诱导剂,在荧光显微镜下观察并记录不同给药组对斑马鱼腹部血管血流停止时间的影响。结果白芍总样、石油醚萃取物、水层样品、芍药苷、白芍苷和芍药苷代谢素-I均表现出抑制血栓生成、促进血液流动的活性;正丁醇相、乙酸乙酯相和芍药苷代谢素-I能够明显增加具有血流的节间血管数量。结论芍药苷代谢素-I具有显著的抑制血栓生成、促进血液流动的活性且优于总样和芍药苷。Objective In order to clarify the medicinal material base and action mechanism,the effect of Radix paeoniae alba extract,major constituents,and metabolite were tested on angiogenesis and antithrombosis in zebrafish. Methods The Radix paeoniae alba were extracted with 70% ethanol and concentrated,and then extracted with petroleum ether,ethyl acetate and n-butanol successively. Healthy transgenic TG(VEGFR2: GFP) zebrafish was used as animal model. Results The growth rate of ISV was recorded to measure the effect of angiogenesis. The result showed that ethyl acetate extract,n-butanol extract and paeonimetabolin I have obvious function of regeneration on ISV of zebrafish; Iron trichloride-induced thrombus significantly improved by total sample,petroleum ether extract,water extract,paeoniflorin,albiflorin and paeonimetabolin I. Conclusion Radix paeoniae alba has the significant effect of angiogenesis and antithrombosis in zebrafish,and the pharmacodynamic material basis may be the metabolite.
