摘 要:【目的】观察腺苷A1受体是否介导参麦注射液对肢体缺血再灌注损伤大鼠的脑保护作用。方法60只大鼠随机分为对照组、模型组、参麦注射液组、参麦注射液+腺苷A1受体拮抗剂组、溶剂对照组。采用双侧股动脉夹闭的方法建立大鼠肢体缺血再灌注损伤模型,对照组大鼠不夹闭股动脉,其他操作同造模大鼠。参麦注射液组大鼠在造模后即刻腹腔注射参麦注射液,模型组大鼠在造模后即刻腹腔注射等量0.9%氯化钠注射液,参麦注射液+腺苷A1受体拮抗剂组大鼠在造模前30 min腹腔注射腺苷A1受体拮抗剂,造模后即刻腹腔注射参麦注射液。溶剂对照组大鼠只在造模前30 min注射二甲亚砜。大鼠肢体缺血再灌注48 h后处死,观察海马组织的脑水含量,采用尼氏染色观察大鼠海马CA1区神经元损伤情况,采用Western blot法观察大鼠海马组织中Bcl-2和Bax蛋白表达量。结果与对照组比较,模型组大鼠的海马组织脑水含量明显增高(P<0.05),Bcl-2/Bax比率明显减小,海马CA1区正常细胞数量明显减少(P<0.05)。与模型组比较,参麦注射液组大鼠海马组织脑水含量明显减少(P<0.05),海马CA1区正常细胞数量明显增多,Bcl-2/Bax比率明显升高(P<0.05)。与参麦注射液组比较,参麦注射液+腺苷A1受体拮抗剂组大鼠海马组织脑水含量明显增加,Bcl-2/Bax比率明显降低,海马CA1区正常细胞数量明显减少(P<0.05)。结论参麦注射液对肢体缺血再灌注损伤大鼠海马组织具有保护作用;腺苷A1受体可能介导了参麦注射液对肢体缺血再灌注损伤大鼠海马组织的保护作用。Objective: To observe the protective effect of Adenosine A1 Receptor mediated Shenmai Injection on hippocampus in rats with limb ischemia-reperfusion injury. Methods: Sixty rats were randomly divided into the control group,the model group,Shenmai Injection group,Shenmai Injection+A1 R antagonist group,the solvent control group,12 in each group. The model was made by a method that the bilateral femoral arteries were occluded with atraumatic microclips. The rats in the control group weren′ t occluded,and the other operation was the same as the model group. Shenmai Injection(15 m L/kg) was peritoneally injected to mice in Shenmai Injection group immediately after models established. Equal volume of normal saline was injected to mice in the model group. A1 R antagonist(DPCPX,1 mg/kg) was peritoneally injected to mice in the Shenmai Injection+A1 R antagonist group 30 min before modeling. DMSO(1 m L/kg) were peritoneally injected to mice in the DMSO control group 30 min before modeling. After limb ischemia-reperfusion 48 h,the mice were killed;water content of hippocampal tissue was detected;the damage of neurons in CA1 area of rat hippocampus was observed by Nissl′ s staining,and the expression of Bcl-2 and Bax protein in rat hippocampal tissues was observed by Western blot method. Results: Compared with the control group,water content of hippocampal tissue was increased(P 0.05);the ratio of Bcl-2/Bax and the normal hippoeampal neurons in CA1 area was decreased in the model group(P 0.05). Compared with the model group,water content of hippocampal tissue was decreased(P 0.05);the ratio of Bcl-2/Bax and the normal hippoeampal neurons in CA1 area was increased in Shenmai Injection group. Compared with Shenmai Injection group,water content of hippocampal tissue was increased;the ratio of bcl-2/bax and the normal hippoeampal neurons in CA1 area was decreased in Shenmai Injection+A1 R antagonist group(P 0.05).Conclusions: Shenmai Injection has protective effects on hippocampal t
