摘  要:细胞凋亡在保证多细胞生物生长发育过程中扮演着重要角色,其在医学领域也有很大的发展空间,对于肿瘤等疾病可以通过诱导突变细胞自主凋亡的思路来治愈疾病。在细胞凋亡过程,抗凋亡因子具有重要的调控作用,因此靶向这些分子可以人为干预细胞凋亡。本研究设计了利用计算机辅助药物设计的方法发现促细胞凋亡药物分子的研究方案,在广泛调研文献的基础上,了解抗凋亡因子Bcl-2家族、p53、c-FLIP,P13K-AKT参与细胞凋亡过程的作用机理,筛选出候选药物靶标,结合计算机辅助药物的方法寻找潜在药物分子,再运用流式细胞仪检测细胞凋亡率,最终筛选出促细胞凋亡的最佳药物分子,该方案为之后的实验和理论研究奠定基础,帮助探索治疗恶性肿瘤等疾病的可能方法。Cell apoptosis plays an important role in ensuring the growth and development process of multicellular organisms,and it also has a great room for development in the field of medicine.For diseases such as tumors,the idea of inducing mutant cells to apoptosis autonomously can be used to cure diseases.Anti-apoptotic factors play an important regulating role in the process of cell apoptosis,so targeting these molecules can interfere with cell apoptosis artificially.This study designed a research plan to discover apoptosis-promoting drug molecules by using computer-aided drug design method.Based on extensive research literature,the mechanism of anti-apoptotic factors Bcl-2 family,p53、c-FLIP,P13 K-AKT participating in the apoptosis process was learnt,candidate drug targets were screened,potential drug molecules were searched by combining computer-aided drug methods,and then the apoptosis rate was detected by using flow cytometry,finally the best drug molecules for promoting cell apoptosis were screened.This plan laid the foundation for subsequent experimental and theoretical research and helped to explore possible methods to treat malignant tumors and other diseases.
