摘 要:目的:研究当归四逆汤对糖尿病血瘀症大鼠周围神经病变及水通道蛋白1(AQP1)、RhoA/ROCK信号通路的影响。方法:65只健康雄性SD大鼠,12只作为正常对照组(A组),其余53只制备2型糖尿病大鼠血瘀症模型。48只成模大鼠再分为:糖尿病血瘀症组(B组)、甲钴胺治疗组(C组)、当归四逆汤低剂量治疗组(D组)和当归四逆汤高剂量治疗组(E组),每组12只。检测各组大鼠坐骨神经传导速度、坐骨神经含水量;背根神经节AQP1和RhoA/ROCK信号通路相关因子mRNA/蛋白表达水平变化。结果:造模大鼠体重明显下降,血糖、血脂、血流变指标水平明显异常(P<0.01);B组大鼠坐骨神经含水量显著高于A组(P<0.01),C组与A组接近(P>0.05),E组与C组相当(P>0.05),好于D组(P<0.05);各组坐骨神经传导速度差异无统计学意义(P>0.05);各组大鼠背根神经节信号通路因子mRNA表达比较,B组AQP1高于A组,RhoA、ROCKII低于A组(P<0.05),C组AQP1、RhoA、ROCKII表达水平接近A组(P>0.05),E组接近C组(P>0.05),D组AQP1表达高于E组,RhoA、ROCKII表达量低于E组(P<0.05),各组ROCKI mRNA表达未见统计学差异(P>0.05);各组大鼠信号通路因子蛋白表达,B组AQP1、RhoA、ROCKII较A组显著下调(P<0.01),C组AQP1、RhoA、ROCKII表达接近A组(P>0.05),E组接近C组(P>0.05),D组表达低于E组(P<0.05),各组ROCKI蛋白表达差异无统计学意义(P>0.05)。结论:当归四逆汤治疗糖尿病周围神经病变有效,且高剂量组效果更好,作用机理可能与其调节AQP1及RhoA/ROCK信号通路表达水平有关。Objective:To study the effect of Danggui si ni decoction on peripheral neuropathy and aquaporin 1(AQP1)and RhoA/ROCK signaling pathway in diabetic rats with blood stasis.Method:65 healthy male SD rats were selected,of these 12 rats as normal control group(group A),the left 53 rats were used for blood stasis syndrome of type 2 diabetic rat model.The 48 model rats were divided into 4 groups:diabetic blood stasis group(group B),Mecobalamin group(group C),angelica si ni decoction low dose group(group D),angelica si ni decoction high dose treatment group(group E).Conduction velocity of sciatic nerve,sciatic content,dorsal root ganglion AQP1 and RhoA/ROCK signaling pathway related factor mRNA and protein expression level were detected.Results:The level of blood glucose,blood lipid and blood rheology indexes in the model group were significantly higher than that of the normal rats(P〈0.05).The water content of the sciatic nerve in the group B was higher than that in the group A and the transmission rate was lower than that of the group A(P〈0.05),and the group C was close to the group A(P〈0.05).The group E was close to group C(P〈0.05)and better than group D(P〈0.05).There was no significant difference in the conduction velocity of sciatic nerve between the two groups(P〈0.05).The mRNA expression of each factor in the dorsal root ganglia of each group was compared.Compared with group A,the AQP1 of group B was higher than that of group A.Compared with group A,the expression levels of AQP1,RhoA and ROCKII in group C were close to group A(P〈0.05),group E was close to group C(P〈0.05),AQP1 expression in group D was higher than group E.The expression of RhoA and ROCKII in group D were lower than that of group E(P〈0.05).The expression of ROCKI mRNA in each group was no significant differencet(P〈0.05).The expression of AQP1,RhoA and POCK II in group D were lower than those of group E(P〈0.05),and those of group E were close to group C(
