摘  要:为建立白芨种子高效实用的组织培养技术体系,以紫花大种白芨成熟未开裂蒴果为外植体,进行组培快繁技术研究。结果表明:外植体表面清洁后,在5%次氯酸钠溶液中浸泡15 min,之后将种子接种于萌发培养基,配方为MS+6-BA(1.0 mg·L^(-1))。种子萌发并形成绿色原球茎即可转入幼苗营养生长培养基,配方为MS+6-BA(2.0 mg·L^(-1))+NAA(0.5 mg·L^(-1))。幼苗生长到3 cm以上时,接入生根培养基中,配方为1/2MS+6-BA(0.1 mg·L^(-1))+NAA(0.5 mg·L^(-1))。待其根系长度达到1 cm后,可进行炼苗移栽。The seed tissue culture system has been preliminarily established by using the mature but not cracking capsule of purple flower Bletilla striata as explant. The basic procedure is as follows. Explants are soaked in 5% HCl O solution for 15 min after surface cleaning,and inoculated on the germinate culture medium until green protocorns formed,the fomula of which is MS + 6-BA( 1. 0 mg·L^-1). The protocorns are subcultured to vegetative growth culture medium until seedling growth over 3 cm,the fomula of which is MS + 6-BA( 2. 0 mg·L^-1) + NAA( 0. 5 mg·L^-1). Then seedlings would be transferred to root medium,the fomula is 1/2 MS + 6-BA( 0. 1 mg·L^-1) +NAA( 0. 5 mg·L^-1). Acclimatization and transplant would be performed after the root growth over 1 cm.
