复方中药洗剂组方的体外抑菌实验研究

摘　　要：目的在复方中药洗剂单药的最佳提取工艺条件下,通过体外抑菌实验确定该洗剂的最优组方及其剂量配比,验证其体外抑菌作用,观察其对阴道常见致病标准菌株的抑菌效果。方法在最佳提取工艺条件下,采用渗漏法提取并干燥白芷、五倍子、川芎、当归、丁香、石斛、人参、珍珠8味单药;选用正交t值法实验设计,以抑菌圈为指标,设定高和低两个剂量水平,观察不同剂量配比的8味单药组成12种组方对白色念珠菌、大肠杆菌、金黄色葡萄球菌、厌氧消化链球菌和阴道加德纳菌5种ATCC标准菌株的体外抑菌作用。结果 12种组方对5种ATCC标准菌株有不同程度的抑菌作用。8味单药对5种ATCC标准菌株的抑菌作用的贡献值都以高剂量为大,除白色念珠菌的珍珠高剂量和低剂量相当,厌氧消化链球菌的人参高剂量和低剂量相当,而阴道加德纳菌的石斛以低剂量为大。12种组方中各单药对5种ATCC标准菌株的抑菌作用,有相加或拮抗作用。结论 12种组方对5种ATCC标准菌株均有一定的抑菌作用,以组方(1)的抑菌效果最强,即原方为最优;组方(1)中以白芷和五倍子为君药(即主药),当归、丁香、石斛、人参和珍珠粉为臣药,川芎为佐使之药。Objective To preliminarily establish the optimal formula and dosage ratio for the detergent,to verify its antibacterial effects in vitro,and to observe the bacteriostatic effect on common vaginal pathogenic standard bacteria by in vitro bacteriostasis experiment under the optimum extraction conditions of Compound Chinese medicine lotion.Methods Under the condition of the optimal extraction process,eight Chinese medicines：dahurian angelica root,nutgall,chuanxiong rhizome,angelica root,clove,dendrobe,ginseng,and pearl were extracted by percolation and then were dried.With Orthogonal intersection of t value,bacteriostatic circle as an index,two levels were set to observe the bacteriostatic effects in vitro of twelve different compositions of the eight medicines on five ATCC standard bacteria：Candida albicans,Escherichia coli,Staphylococcus aureus,Peptostreptococcus anaerobius,and Gardnerella vaginalis.Results The twelve compositions had different degrees of bacteriostatic effects on the five ATCC standard bacteria.The contributions of eight medicines to bacteriostatic effects were all higher at high doses,except the effect of pearl on Candida albicans were equal at low dose and high dose,the effect of ginseng on Peptostreptococcus anaerobius were equal at low dose and high dose,and the effect of dendrobe on Gardnerella vaginalis was higher at low doses.The bacteriostatic effects of the twelve compositions on the five ATCC standard bacteria were enhanced or antagonistic.Conclusion The twelve compositions on the five ATCC standard bacteria all have certain antibacterial effects.Composition（1）is the strongest,i.e.the original prescription.The composition（1）uses dahurian angelica root and nutgall as the main ingredient,supplemented by angelica root,clove,dendrobe,ginseng,pearl powder.Chuanxiong rhizoma is the guiding medicine.
