摘 要:目的探讨甘松挥发油对大鼠心肌缺血再灌注损伤及磷脂酰肌醇3激酶(PI3K)通路的的影响。方法将80只SD大鼠随机分成5组,即假手术组(n=10)、心肌缺血再灌注(IR)组(n=20)、高剂量甘松挥发油(HD)组(n=20)、低剂量甘松挥发油(LD)组(n=20)、二甲基亚砜(DMSO)组(n=10)。观察各组大鼠心肌梗死面积、心肌损伤标记物、心率、血压变化,以及PI3K通路蛋白表达情况。结果高剂量甘松挥发油组大鼠心肌梗死面积、血肌酸激酶同工酶MB(CK-MB)和心肌肌钙蛋白(cTnT)表达均显著小于IR模型组(P<0.05),同时高剂量甘松挥发油组Akt/磷酸化Akt(p-Akt)、GSK-3β/磷酸化GSK-3β(p-GSK-3β)、eNOS/磷酸化eNOS(p-eNOS)蛋白表达显著高于IR模型组(P<0.05)。结论甘松挥发油后处理大鼠心肌缺血再灌注能显著减轻心肌损伤,其保护机制可能与PI3K-AKT通路激活有关。Objective To investigate the effects of the volatile oil of nardostachys chinensis on reperfusion injury and PI3K signaling pathway in myocardial ischemia rats. Methods Eighty SD rats were randomly divided into 5 groups: sham group( n =10); ischemia reperfusion(IR) group ( n = 20); high dose volatile oil of nardostachys Chinensis group ( n =20); low dose volatile oil of nardostachys chinensis(LD) group( n =20); dimethyl sulfoxide (DMSO) group ( n =10). The myocardial infarction size, markers of myocardial injury, changes of heart rate and blood pressure, and the expression of PI3K signaling pathway were observed. Results The serum levels of serum creatine kinase isoenzyme MB (CK-MB), cardiac troponin (cTnT), and myocardial infarction size were significantly less in the high dose volatile oil of nardostachys chinensis group ( P 〈0.05), and the expressions of Akt/phosphorylate Akt(p-Akt), GSK-3β/phosphorylate GSK-3β(p-GSK-3β), eNOS/phosphorylate eNOS(p-eNOS) were higher than IR model group( P 〈0.05). Conclusion Volatile oil of nardostachys chinensis have protective effects on myocardial ischemia/reperfusion injury, and the protective mechanism may be associated with the PI3K signaling pathway.
