摘 要:稻纵卷叶螟是一种主要的水稻害虫,海藻糖酶是昆虫几丁质合成通路上的关键酶之一。该研究对转稻纵卷叶螟可溶性海藻糖酶基因(CmTre1)双链RNA(dsRNA)水稻进行了室内和田间抗虫性鉴定。叶片测定结果表明,取食转CmTre1基因dsRNA水稻株系608的稻纵卷叶螟死亡率为70.00%,校正死亡率为50.95%。定量PCR检测表明,幼虫取食608株系后其体内CmTre1基因的mRNA水平下降59.00%,该基因的表达被抑制。田间抗虫性检测结果表明,608株系的卷叶率为15.06%,白叶率为13.98%,与对照差异显著。转CmTre1基因RNA干扰(RNAi)水稻对稻纵卷叶螟具有抗性。该研究为利用RNAi技术对稻纵卷叶螟进行绿色防控奠定基础。The rice leaf folder,Cnaphalocrocis medinalis,is a major rice pest. Trehalase( Tre) is one of the key enzymes in the chitin synthesis pathway of insects. In this study,we identified the insect resistance of rice that was introduced the double-stranded RNA( dsRNA) of soluble trahalase gene from C. medinalis( CmTre1) indoors and in the fields. Results of leaf assay showed that mortality rate of C. medinalis fed on the dsRNA-transgenic rice line 608 was 70. 00% and the corrected mortality rate was 50. 95%. Real-time quantitative PCR( RT-qPCR) analysis demonstrated that mRNA level of CmTre1 in C. medinalis fed on rice 608 dropped by 69. 00%,and the expression of CmTre1 gene was inhibited. Results of insect resistance test in the fields indicated that the average rate of rolled and white leaves for rice 608 was 15. 06% and 13. 98%,respectively,which was significantly different from the control. The CmTre1 dsRNA-transgenic rice was resistant to C. medinalis. This study would lay the foundation for the green control of C. medinalis using RNA interference( RNAi) technology.
