摘 要:采用双水相萃取技术,研究alpha-酮戊二酸在低分子有机溶剂/无机盐双水相体系中的分配行为.用双光束紫外可见光分光光度计对alpha-酮戊二酸进行定性定量测定,确定了最佳吸收波长为235.14nm,标准曲线为C=1.8239A+0.0128(R2=0.999),在0.005~0.250g/L范围内呈良好的线性关系.采用亲水性有机溶剂丙酮/K_2HPO_4·3H_2O的新型双水相体系,研究丙酮用量、K_2HPO_4·3H_2O用量以及萃取温度对α-酮戊二酸在两相中分配的影响,确定了最佳工艺条件:系统总量为40g,丙酮用量为9.6g,K_2HPO_4·3H_2O用量为6g,萃取温度为30℃.在此条件下,alpha-酮戊二酸的分配系数K=2.5,萃取率E=87.56%.The distribution behavior of alpha-ketoglutarate in aqueous two-phase systems containing low molecular organic solvent and inorganic salt was studied.Double beam ultraviolet-visible spectrophotometer was used for the qualitative and quantitative determination of alpha-ketoglutarate.The best absorption wavelength was 235.14 nm and the standard curve was C=1.8239 A+0.0128(R2=0.999)in range of 0.005~0.250(g/L).In this experiment,a new aqueous two-phase system contained hydrophilic organic solvent acetone and dipotassium hydrogen phosphate was adopted.The effects of acetone dosage,dipotassium hydrogen phosphate dosage and extraction temperature on the distribution of alpha-ketoglutarate in two phases were studied.The optimum technological conditions were determined:the system total amount was 40 g,acetone dosage was 9.6 g,dipotassium hydrogen phosphate dosage was 6 g,extraction temperature was 30℃.In this condition,the allocation coefficient of alpha-ketoglutaric acid was K= 2.5,and the extraction rate was E=87.56%.
