摘  要:目的探讨大鼠肝移植后肝再生增强因子(augmenter of liver regeneration,ALR)与移植物急性排斥反应的关系。方法建立急性排斥组及免疫耐受组大鼠肝移植动物模型各12只。术后第1、3、5和7天取大鼠外周血浆及肝脏组织标本。检测外周血浆中丙氨酸氨基转移酶(ALT)及天冬氨酸转移酶(AST)和总胆红素(TBIL)水平;检测肝脏移植物组织内ALR、干扰素-γ(INF-γ)和白细胞介素-2(IL-2)的表达;分析ALR与INF-γ和IL-2表达变化关系。结果术后第1天急性排斥组INF-γ明显高于免疫耐受组,第3、5、7天两组大鼠外周血ALT、AST及TBIL均高于术后第1天,且急性排斥组INF-γ、IL-2表达及ALR的表达明显高于免疫耐受组P<0.05)。相关性分析发现肝移植急性排斥过程中ALR和INF-γ的表达呈明显的负相关(P<0.05),而与IL-2的表达无明显相关性(P>0.05)。结论 ALR与大鼠肝移植后急性免疫排斥反应呈明显的负相关,可能参与了这一过程的调控。Objective To investigate the correlation between expression of augmenter of liver regeneration( ALR) and acute rejection after rat orthotopic liver transplantation.Methods We established rat models of acute rejection and immune tolerance,respectively.Twelve rats in each group.Serum and liver tissue were collected on 1,3,5 and 7 days after surgery. Serum ALT,AST and TBIL were detected by biochemical analysis.The intra-graft expressions of ALR,INF-γ and IL-2 were evaluated to analyze the relationship between ALR and INF-γ/IL-2.Results INF-γ in the acute rejection group was significantly elevated compared to the immune tolerance group on the first day after orthotopic liver transplantation.ALT,AST and TBIL were gradually elevated in the both groups on days 3,5 and 7 after transplantation when compared to the first day.The expressions of INF-γ and IL-2 in the acute rejection group were significantly higher than those in the immune tolerance group( P〈0. 05).Correlation analysis showed that there was a negative correlation between ALR and INF-γ( P〈0. 05) while there was no correlation between ALR and IL-2( P〈0. 05) during the process of the acute rejection.Conclusion ALR is negatively correlated with acute rejection after rat allogenic liver transplantation,suggesting that ALR may be participated in regulating acute rejection.
