摘  要:目的采用UPLC-MS/MS法测定对乙酰氨基酚栓中对乙酰氨基酚的含量。方法色谱柱:ACQUITYTM UPLC BEH C18(2.1mm×50mm,1.7μm);流动相:甲醇-5mmol·L^(-1)乙酸铵(含2mL·L^(-1)乙酸)(65∶35);流速:0.2mL·min-1;进样量:2μL;柱温:30℃,质谱采用ESI+多反应监测模式(MRM)进行扫描。结果对乙酰氨基酚在3min内分离良好,在相应的质量浓度范围内与峰面积呈良好的线性关系(r=0.995 7,n=6);加样回收率为99.2%(n=6),RSD值为2.0%。结论建立的UPLCMS/MS分析方法简便、快速、灵敏度高、专属性强、结果准确,适用于对乙酰氨基酚栓的质量控制。Objective To determine the content of acetaminophen in Paracetamol Suppositories by UPLC-MS/MS.Methods The LC conditions were as follows:the column,ACQUITYTM UPLC BEH C_(18)(2.1 mm×50 mm,1.7μm);the mobile phase,methanol-5 mmol·L^(-1) ammonium acetate(containing 2 mL·L^(-1) acetic acid)(65∶35);the flow rate was 0.2 mL·min^(-1);the injection volume was 2μL;the column temperature was 30 ℃.The mass spectra were recorded by using ESI+multiple reaction monitoring(MRM).Results Acetaminophen was well separated within 3 min and had a good linear relationship with the peak area in the corresponding concentration range(r=0.995 7,n=6).The recovery was 99.2%(n=6),and RSD was 2.0%.Conclusion The established UPLC-MS/MS method is simple,rapid,sensitive,specific and accurate.It is suitable for the quality control of Paracetamol Suppositories.
