肠激安制剂对大鼠的长期毒性实验研究

摘　　要：目的观察肠激安制剂连续灌胃给药对大鼠所产生的毒性反应,探讨肠激安制剂临床用药安全性。方法肠激安制剂分别按照高(15.624g·kg^(-1))、中(7.812g·kg^(-1))和低(3.906g·kg^(-1))剂量给药,连续灌服6周,停药2周。每周测定大鼠体质量,进食量以及给药6周和停药2周后各组大鼠的脏器指数、尿常规、血常规和血生化指标,并做组织病理学检查。结果与空白对照组比较,口服肠激安制剂高、中、低剂量6周对大鼠的体质量增长、进食量、脏器系数、血液指标和血液生化学指标均无显著影响,病理学检查未见明显病变,停药后也未见延迟性毒性反应。结论肠激安制剂长期用药对大鼠无明显毒性,临床拟用剂量安全性良好。Objective To research the chronic toxicity of Changji′an Preparation in rats and to discuss the safety of clinical medication of the preparation.Methods The rats were orally given for 6 weeks with the doses of 15.624,7.812 and 3.906 g·kg（-1）.After stopping administration 2 weeks,the weights,organ coefficients,the indexes of hematology and biochemistry was determined,and the pathological examination was carried out.Results The outer appearance and behavior,weights,organ coefficients,the indexes of hematology and biochemistry showed no significant difference by comparing with control group;no significant pathological changes with relation to drug toxicity and the delayed toxicity reaction in stopping administration later was found by pathological examination.Conclusion Changji′an Preparation has no significant toxicity for long-term administration in rats,the proposal dose in clinic is suitable.
