摘 要:朊蛋白因在一类称为朊病毒病的神经退行性疾病中扮演着重要角色而得到了广泛研究。目前,虽然在病理和传播方面已对这类疾病开展了大量研究,但致病的分子机制仍未阐明。那么,如何理解朊病毒病中朊蛋白致病的分子机制呢?从朊蛋白的生理功能中可以获得哪些启发呢?伴随着这些问题,相信深入了解朊蛋白的生理功能可揭示其参与朊病毒病的致病过程。朊蛋白已被证明参与了许多生物学过程,如神经突触生成、神经内稳态、细胞信号转导、细胞粘附和抗应激的保护性作用。朊蛋白的这种多能性使研究人员无法准确了解朊蛋白的分子功能。将努力阐明朊蛋白在中枢神经系统和外周神经系统中的生理作用。The prion protein( Pr P) has been extensively studied because of its central role in a group of neurodegenerative conditions collectively known as prion diseases. While the wealth of information was available for the pathology and transmission of these diseases,the molecular mechanisms involved were not yet clearly defined. So,how to learn about the molecular mechanisms for the pathogenic role of the Pr P in disease,and get enlightenment about the physiological role of the Pr P Defining Pr P function might illuminate pathological processes involved in prion diseases. The Pr P had been shown to participate in several biological processes including neuritogenesis,neuronal homeostasis,cell signalling,cell adhesion,and a protective role against stress. This pleiotropism had been led to confusion about the precise molecular function( s) of the Pr P. This essay should attempt to clarify the most relevant physiological roles of the protein in the context of the central and peripheral nervous system.
