摘  要:背景:有研究证实在电刺激条件下,聚苯胺/聚乳酸复合支架可促进神经干细胞突起的生长。目的:制备电活性聚苯胺/聚乳酸支架,观察其与大鼠嗅鞘细胞的生物相容性。方法:采用乳液聚合的方法制备聚苯胺/聚乳酸膜,电镜下观察其形态,测定其电导率。将SD乳鼠嗅鞘细胞分别接种于聚苯胺/聚乳酸膜上(实验组)与多聚赖氨酸包被的玻片上(对照组),进行细胞增殖、细胞黏附率测定及免疫荧光抗体标记观察。结果与结论:(1)扫描电镜下,聚苯胺/聚乳酸膜表面平整、均匀,可见为细小纤维形成的网格状结构,其电导率为1.5×10-5 S/cm;(2)培养1,3,5,7 d,实验组的细胞相对增殖率均在90%以上,细胞毒性为Ⅰ级;(3)培养7 d后,实验组细胞吸附率高于对照组(P<0.05);(4)培养7 d后免疫荧光检测显示,对照组S-100阳性细胞的数量及胞体面积与实验组无明显性差异(P>0.05),对照组S-100阳性细胞周长明显小于实验组(P<0.05);(5)结果表明,制备的聚苯胺/聚乳酸膜与嗅鞘细胞具有良好的生物相容性。BACKGROUND: Previous studies have confirmed that polyaniline/poly(lactic-acid) scaffold could promote the growth of neural stem cell processes in the condition of electrical stimulation.OBJECTIVE: To prepare an electroactive polyaniline/poly(lactic-acid) scaffold and to investigate its biocompatibility with rat olfactory ensheathing cells. METHODS: Olfactory ensheathing cells of Sprague-Dawley neonatal rats were inoculated onto the polyaniline/poly(lactic acid) film(experimental group) and poly lysine coating slides(control group). Cell proliferation and cell adhesion rate were detected, and the cells were observed with immunofluorescence histochemistry. RESULTS AND CONCLUSION:(1) Smooth and even polyaniline/poly(lactic acid) film was visible under electron microscopy, with a grid structure formed by fine fibers, and its conductivity was 1.5×10-5 S/cm.(2) At 1, 3, 5, 7 days after co-culture, the cell relative growth rates were all above 90%, and the cytotoxicity was graded I.(3) At 7 days after co-culture, the cell adhesion rate in the experimental group was significantly higher than that in the control group(P〈0.05).(4) At 7 days after co-culture, immunofluorescence results showed that there was no obvious difference between the two groups in the number or area of S-100 positive cells(P〉0.05), while the cell perimeter in the experimental group was significantly higher than that in the control group(P〈0.05). To conclude, the prepared polyaniline/poly(lactic acid) film has good biocompatibility with olfactory ensheathing cells.
