摘  要:目的分析姜黄素减轻过氧化氢诱导皮层神经细胞凋亡的JAK信号通路机制,为相关新药开发及临床治疗提供参考。方法在37℃、5%CO2条件下体外培养胎鼠大脑皮层神经细胞至生长良好。将皮层神经细胞随机分为四组:对照组、过氧化氢组、姜黄素组及S1491组。姜黄素组(40μmol/L)及S1491(20μmol/L)组细胞预先给予相应浓度药物预处理4 h,对照组给予生理盐水处理。然后除对照组给予生理盐水外,其余各组细胞给予400μmol/L的过氧化氢共培养8 h。分析各组细胞经相应的处理后细胞的增殖抑制率,采用q PCR法和WB法分析各组皮层神经细胞Caspase3、Caspase6、JAK通路蛋白水平表达。结果与对照组相比,过氧化氢组细胞增殖被明显抑制(P<0.05),细胞凋亡蛋白Caspase3、Caspase6及JAK通路蛋白JAK1和JAK2表达明显增强(P<0.05),而姜黄素组及S1491组细胞的上述异常得到明显地恢复(P<0.05)。结论姜黄素能有效减轻过氧化氢诱导皮层神经细胞凋亡过程,且其可能是通过抑制JAK信号转导通路发挥作用。Objective To analyze the JAK signaling pathway that curcumin alleviated the apoptosis of cortical neurons induced by hydrogen peroxide. Methods Under the condition of 37℃ and 5%CO 2, the cultured fetal rat cerebral cortex nerve cells grew well in vitro. The cortical neurons were randomly divided into four groups: the control group, the hydrogen peroxide group, the curcumin group and the S1491 group. The cells in the curcumin group (40 μmol/L) and the S1491 (20 μmol/L) group were pretreated with the corresponding concentration for 4 h. The control group was treated with saline. In addition, the control group received saline, the other groups were given 400 μmol/L of hydrogen peroxide to co-cultured 8 h. The cell proliferation inhibition rate of each group of cells after treatment was analyzed. The expression of Caspase3, Caspase6 and JAK pathway proteins in cortical neurons of each group was analyzed by qPCR and WB methods. Results Compared with the control group, the proliferation of H 2O 2 group was significantly inhibited ( P 〈0.05), and the expression of apoptotic protein Caspase3, Caspase6 and JAK pathway protein JAK1 and JAK2 increased significantly ( P 〈0.05). The above abnormality of the curcumin group and the S1491 group cells were obviously recovered ( P 〈0.05). Conclusion Curcumin can effectively alleviate the apoptosis of cortical neurons induced by hydrogen peroxide, and it may play a role by inhibiting the JAK signal transduction pathway.
