摘  要:目的研究维生素D受体(VDR)基因启动子甲基化异常与婴儿尿路感染的关系。方法选择急性尿路感染患儿32例(尿路感染组)和同期健康体检婴儿30例(健康对照组)。采用荧光定量PCR法检测VDR、DNA甲基化转移酶(DNMTs)m RNA表达水平,重亚硫酸氢钠测序技术检测VDR基因启动子的甲基化水平。结果尿路感染组患儿VDR m RNA表达水平高于健康对照组婴儿,DNMT1 m RNA表达水平低于健康对照组婴儿,两组比较差异均有统计学意义(均P<0.05)。两组DNMT3a、DNMT3b m RNA表达水平比较差异均无统计学意义(均P>0.05)。尿路感染组患儿VDR基因启动子甲基化水平显著低于健康对照组婴儿,两组比较差异有统计学意义(P<0.05)。结论尿路感染患儿VDR m RNA表达水平升高,VDR基因启动子的甲基化水平降低,VDR基因甲基化异常与婴儿尿路感染相关。Objective To analyze the relationship between methylation of vitamin D receptor(VDR) gene promoters and urinary tract infection(UTI) in infants. Methods Thirty two infants aged 1 ~12 months with UTI(UTI group) and 30 normal infants with matched ages(control group) were enrolled in the study. The mRNA expressions of VDR and DNA methyltransferase(DNMT1, DNMT3a, DNMT3b) were detected by real-time PCR; the methylation levels of VDR gene promoters were detected by bisulfite sequencing(BSP). Results The expression of VDR mRNA in UTI group was higher than that in control group(P〈0.05). The value of DNMT1 mRNA in UTI group was lower than that in control group(P〈0.05). There was no significant difference in expression levels of DNMT3a mRNA and DNMT3b mRNA between two groups(both P〈0.05). The methylation of VDR gene promoter in UTI group was significantly lower than that in control group(P〈0.05). Conclusion The expression level of VDR mRNA is up-regulated in patients with UTI, while the methylation of VDR gene promoter is decreased, abnormal methylation of VDR gene is associated with urinary tract infection in infants.
