摘 要:根据GB/T 15000.3–2008研制岩藻黄醇标准样品。以岩藻黄质提取物为原料,通过酶解、提取、柱层析、制备分离获得高纯岩藻黄醇单体,采用红外光谱、高分辨质谱和核磁共振波谱等方法进行结构确证。将样品分装为150瓶,随机抽取15个样品,采用高效液相色谱–紫外检测法进行稳定性、均匀性检验。检验结果表明,在95%的置信区间范围内本样品均匀性良好,于–18℃条件下保存12个月,其稳定性良好。经8家具有分析资质的实验室进行协同定值,岩藻黄醇标准样品特性标准值(纯度)为99.35%,相对扩展不确定度为0.49%(k=1.96)。研制的岩藻黄醇标准样品可用于岩藻黄醇相关产品的检验和质量控制。Fucoxanthinol reference material was developed according to GB/T 15000.3–2008. Fucoxanthinol was made from the Fucoxanthin. High purity fucoxanthinol monomer was obtained from the extracting of fucoxanthin by enzymolysis, extraction, column chromatography and separation. The structure of fucoxanthinol certified reference material was confirmed by infrared spectroscopy,high resolution mass spectral and nuclear magnetic resonance method. Fucoxanthinol samples was divided into 150 bottles, and 15 bottles of samples were randomly taken. The homogeneity and stability testing were carried out by high performance liquid chromatography–ultraviolet detector. The detection results indicated that homogeneity and stability of the fucoxanthinol reference material were well, the period of fucoxanthinol storage was 12 months at –18℃ . A cooperative certification was conducted by 8 laboratories with analytical qualifications. The certified value(purity) of the reference material of fucoxanthinol was 99.35% with relative expaned uncertainty of 0.49%(k=1.96). The developed reference material of fucoxanthinol can be used for detection and quality control of fucoxanthinol related products.
