摘  要:目的研究ADAM33基因ST+4位点多态性与中国黑龙江省儿童哮喘的发生及其严重程度是否存在关联性。方法采用聚合酶链反应(PCR)及DNA测序方法,对105例儿童哮喘患者及98例健康儿童ADAM33基因ST+4位点多态性进行检测分析,并计算其基因型频率及等位基因频率,用SPSS17.0进行数据处理。结果 ADAM33基因ST+4位点3种基因型(CC,CA,AA),其中CC基因型在病例组患儿中的频率明显高于对照组(χ~2=7.22 P=0.007),而病例组AC基因型频率明显低于对照组(χ~2=6.56 P=0.01),AA基因型频率在病例组及对照组中无明显差异(χ~2=0.03 P=0.86),提示CC基因型可能增加了中国东北地区儿童哮喘的危险性,AC基因型可能是儿童哮喘的保护因素。等位基因C的频率在病例组中明显高于对照组,C及A的等位基因频率具有显著差异(χ~2=4.29 P=0.038),提示C等位基因增加了儿童哮喘的易感性。将病例组分为轻,中,重3组,分别与对照组相比较,而发现ST+4位点与重度哮喘有关(P<0.01)。进一步验证,在轻,中,重度哮喘之间进行对比发现,轻中度哮喘与重度哮喘之间存在差异(P<0.01)。结论 ADAM33基因ST+4位点与中国东北地区儿童重度哮喘有关。Objective To study the relevance between the ADAM33 gene SNP ST + 4 and susceptibility and severity of pediatric asthma in Heilongjiang province children. Methods The ADAM33 gene SNP ST + 4 of 105 pediatric asthma patients and 98 healthy children were detected by PCR and DNA sequencing methods. The data were analyzed by SPSS 17. 0. Results The ADAM33 gene SNP ST + 4 CC genotype frequency in the asthmatic group was obviously higher than that in the control group( χ^2= 7. 22,P = 0. 007). Among the asthmatic subjects,AC genotype frequency was lower than healthy controls( χ^2= 6. 56,P = 0. 01). There was no significant difference in AA genotype frequency between the two groups( χ^2= 0. 03,P = 0. 86). The results revealed CC genotype might increase the risk of asthma,and the AA genotype might decrease the risk of asthma in Northern Chinese children. The frequencies of alleles( C,A) were also different between the two groups( χ^2= 4. 29,P = 0. 038),and the C alleles was dangerous for prediatric asthma. The asthmatic group was divided three different groups,including the mild,moderate,severe asthma group. While compared with healthy controls,SNP ST + 4 had a significant relationship with severe asthma( P〈0. 01). For further study,the results revealed mild and moderate asthma had a difference with severe asthma( P〈0. 01). Conclusion ADAM33 gene SNP ST + 4 is associated with severe pediatric asthma in Heilongjiang province children.
