摘 要:目的建立了高效液相色谱法同时测定云南6个产地滇丹参中5种水溶性成分含量,为滇丹参资源的合理开发利用提供依据。方法采用Agilent 1200系列高效液相色谱仪,Agilent ZORBAX Eclipse XDB-C_(18)色谱柱(4.6 mm×150 mm,5μm),乙腈-0.2%磷酸水溶液为流动相进行梯度洗脱,流速1.0 mL/min,检测波长280nm,柱温25℃。结果 5种水溶性成分均呈现较好的回归方程和相关系数,原儿茶醛、咖啡酸、紫草酸相关系数均达到0.9999,丹酚酸B为0.9998,丹参素为0.9969。其中丹参素进样范围为(6.08~39.52)×10^(-2)μg,原儿茶醛进样范围为(3.23~21.01)×10^(-3)μg,咖啡酸进样范围为(8.56~55.64)×10^(-3)μg,紫草酸进样范围为(32.56~211.64)×10^(-2)μg,丹酚酸B进样范围为(102.48~666.12)×10^(-2)μg,丹参素平均回收率为99.353%,原儿茶醛平均回收率为99.077%,咖啡酸平均回收率为99.023%,紫草酸平均回收率为98.750%,丹酚酸B平均回收率为99.098%。结论云南6个产地滇丹参药材中的水溶性成分含量有较大差异,其中新平和洱源产的药材水溶性成分总含量明显高于其他产地,丹酚酸B是这5种水溶性成分中的主成分。本检测方法简单准确、高效、稳定性好,可为滇丹参质量控制及地方药材资源的充分开发利用提供参考依据。Objective To establish a method for determination of 5 kinds of hydrophillic components in Salvia yunnanensis C.H.Wright from six regions in Yunnan by HPLC,to provide the basis for the rational exploitation and utilization of Salvia yunnanensis C.H.Wright.Methods The Agilent 1200 series high performance liquid chromatography was used,Agilent ZORBAX Eclipse XDB-C18 column (4.6mm×150mm,5μm)was chosen,acetonitrile-0.2% aqueous solution were used as gradient elution for mobile phase,the flow rate was 1 mL/min,the detection wavelengh was 280nm,and the column temperature was 25 degrees.Results 5 kinds of hydrophillic components showed good regression equation and correlation coefficient.The correlation coefficients of protocatechuic aldehyde,caffeic acid and purple oxalate reached 0.9999,salvianolic acid B was 0.9998 and danshensu was 0.9969.The sampling range of salvianic acid was (6.08~39.52)×10-2 μg,protocatechuic aldehyde was (3.23~21.01)×10-3 μg,caffeic acid sample was (8.56~55.64)×10-3 μg,alkannic acid was (32.56~211.64)×10-2 μg,salvianolic recovery rate of protocatechualdehyde was 99.077%,the average recovery rate of caffeic acid was 99.023%,the average recovery rate of alkannic acid was 98.750% alkannic,the averages recovery rate of salvianolic acid B was 99.098%.Conclusion Hydrophillic components of Salvia miltiorrhiza in 6 origin from Yunnan have great difference,the total content of hydrophillic components from Xinping and Eryuan is significantly higher than that in other areas.Salvianolic acid B is the principal component of these 5 hydrophillic components.This method is simple,accurate,efficient and stable,which can provide reference for the quality control of Salvia yunnanensis C.H.Wright and the full exploitation and utilization of local medicinal materials.
