中药肾精子对大鼠前列腺增生模型的影响研究

摘　　要：目的:通过研究肾精子对大鼠前列腺增生模型代谢尿量、前列腺脏器指数、激素水平、前列腺病理改变等的影响,验证肾精子利水消肿的功效。方法:采用雄性大鼠去势同时给予定量丙酸睾酮的方法复制大鼠前列腺增生模型,给予肾精子进行干预,采用大鼠5 h水负荷代谢尿量、前列腺脏器指数、血清酸性磷酸酶(PAP)、睾酮(T)及雌二醇(E2)、前列腺病理组织改变等作为评价指标进行评价。结果:给药第4周5 h水负荷代谢尿量模型组与空白组具有显著统计学意义(P<0.05),模型组与给药组比较具有统计学意义(P<0.05);前列腺脏器指数模型组与空白组比较有显著统计学意义(P<0.01),肾精子给药组与模型组比较具有统计学意义(P<0.05);模型组大鼠前列腺组织增生严重,肾精子给药组前列腺组织部分增生;模型组血清中PAP、T、E2水平明显升高,给予肾精子后能有效降低血清中PAP、T、E2水平。结论:5 h水负荷代谢尿量可作为大鼠前列腺增生模型指标,肾精子可有效抑制前列腺细胞增生,对前列腺增生症有较好的药效作用。Objective：To research the influences of traditional Chinese drug Shenjingzi on metabolic urine volume,prostate organ indexes,hormone level and change of prostatic pathology etc.of prostatic hyperplasia model of rat,and to observe the efficacy of Shenjingzi on inducing diuresis for removing edema.Methods：By castrating male rats and simultaneously administrating quantitative testosterone propionate,the prostatic hyperplasia models of rats was duplicated.The models were intervened with Shenjingzi and evaluated by metabolic urine volume of 5-hour water load,prostate organ indexes,PAP,T,E2 and change of prostatic pathological tissue etc.evaluation indexes.Results：After 4 weeks of administration,in metabolic urine volume of 5-hour water load,compared model group with blank group,there was a statistically significant difference（P〈0.05）;compared model group with administration group,there was a statistically significant difference（P〈0.05）.In prostate organ indexes,compared model group with blank group,there was a statistically significant difference（P〈0.01）;compared the administration group of Shenjingzi with model group,there was a statistically difference（P〈0.05）.In the model group,the prostatic tissue hyperplasia of rats was severe.In the administration group of Shenjingzi,the prostatic tissue was partially hyperplastic.In serum of model group,the levels of PAP,T and E2 increased significantly.After administration of Shenjingzi,the levels of PAP,T and E2 in serum were effectively reduced.Conclusion：The metabolic urine volume of 5-hour water load can be used as an index of prostatic hyperplasia model of rat.Shenjingzi can effectively inhibit the proliferation of prostate cells,and has a better effect on prostatic hyperplasia symptom.
