泰山白首乌、赤首乌及市售何首乌中游离大黄素和大黄素甲醚的比较研究

摘  要:目的采用高效液相色谱法测定白首乌、赤首乌以及市售何首乌中大黄素、大黄素甲醚的含量,旨在比较不同种类何首乌中两种成分含量的差异。方法采用大连依利特Sinochom ODS-BP色谱柱(4.6 mm×200 mm,5μm),以甲醇-0.1%磷酸溶液(90︰10)为流动相,检测波长254 nm,流速1.0 ml/min,进样量20μl,柱温25℃。结果大黄素、大黄素甲醚标准品在1.0~25μg/ml的浓度范围内,线性关系良好;泰山白首乌中未检测到大黄素及大黄素甲醚;赤首乌中大黄素和大黄素甲醚的含量分别为:33.49μg/g、35.21μg/g;市售何首乌中大黄素和大黄素甲醚的含量分别为88.03μg/g、33.43μg/g。结论本方法快速、准确,可用于何首乌中大黄素和大黄素甲醚的含量测定;不同种类何首乌中大黄素和大黄素甲醚的含量存在差异。Objective: Determinating the content of emodin and physcion in different kinds of Fallopia multiflorum Thunb by HPLC method,aims to compare the difference. Methods: The analyses were performed on a Elite Sinochom ODSBP column( 4. 6 mm × 250 mm,5 μm),and the mobile phase was methanol-water containing 0. 1% phosphoric acid( 90 ︰10) at the flow rate of 1 ml/min. The detection was carried out at 245 nm,the sample size is 20μl,and the column temperature was 25 ℃. Results: Emodinin and physcion in the concentration range of 1. 0 - 25 μg/ml,has a good linear relationship; The content of emodin and physcion in Fallopia multiflorum Thunb is 33. 49 μg/g,35. 21 μg/g; in the Fallopia multiflorum Thunb from market is 88. 03 μg/g,33. 43 μg/g; and in the radix cynanchi Auriculati does not detect the emodin and physcion. Conclusion: This content determination method is accruate,fast,easy to operate,has good stability,and can be used to be determine the content of emodin and Physcion; there has differences between different Fallopia multiflorum Thunb.
