摘 要:目的蔗糖合酶(Sucrose Synthase)是植物蔗糖代谢途径中的关键酶,在植物的生长发育过程中具有重要作用。分析蔗糖合酶的核酸序列信息,预测其蛋白结构与功能,对进一步揭示该酶生物学功能具有重要指导意义。方法将从白及转录组数据库中筛选得到蔗糖合酶核酸序列,采用生物信息学软件进行全长核酸序列信息与氨基酸结构分析。结果该基因全长2 957 bp,编码816氨基酸。蛋白结构预测表明分子量为93.09 k D,理论等电点(p I)为5.97。该蛋白质二级结构主要为α-螺旋结构和无规则卷曲结构,结构域分析则发现该蛋白含有蔗糖合酶功能域和糖基转移功能域。同时,该蛋白与铁皮石斛氨基酸序列高度相似,达91.54%。以其与15种近源物种的蔗糖合酶氨基酸序列构建进化树,分析表明白及与铁皮石斛在进化关系上最近。结论该基因具有典型蔗糖合酶的结构特征,氨基酸序列及核酸序列与同源物种相似度高,在进化上高度保守。Objective To analyze the nucleic acid sequences information of sucrose synthase( Su Sy),the key enzyme in Sucrose metabolism pathway,and predict the protein structure and further reveal the biological function of the enzyme. Methods After Su Sy sequence was selected from transcriptome database of B. striata,a series of online bioinformatics tools was used to analyze the queries. Results The B. striata Su Sy was 2 957 bp in length,coding polypeptides of 816 amino acids with a calculated molecular mass of 93. 09 k D and an isoelectronic point of 5.97. The secondary structure of Su Sy was mainly composed with alpha helixes and random coils,with Su Sy and glycos transf 1 conserved domain. At the same time,the Su Sy was highly similar to the amino acid sequence of Dendrobium candidum with 91. 54% similarity. Phylogenetic analysis based on its Su Sy amino acid sequence with its15 near-origin species revealed that the similarity relationship between B. striata and D. candidum was the closest. Conclusion The gene has the structural characteristics of a typical Su Sy. The amino acid sequence and the nucleic acid sequence have high similarity with the homologous species and are highly conserved.
