摘  要:目的考察通络醒脑泡腾片对血管性痴呆(VD)模型大鼠视交叉上核生物钟基因表达的影响。方法采用双侧结扎颈总动脉法(2-VO法)制备血管性痴呆大鼠模型,采用Morris水迷宫评价其认知功能;反转录聚合酶链式反应(RT-PCR)法检测各组大鼠视交叉上核生物钟基因Clock、Bmal、Per2基因的表达。结果通络醒脑泡腾片能够改善血管性痴呆模型大鼠学习记忆功能,可上调模型大鼠视交叉上核Bmal、Clock和Per2基因表达(P>0.05)。结论通络醒脑泡腾片调节血管性痴呆模型大鼠认知能力与其调节视交叉上核生物钟基因Bmal、Clock和Per1有关。Objective To explore the effect of Tongluo Xingnao Effervescent Tablets on the expression of Clock,Bmal and Per2 genes in the suprachiasmatic nucleus of VD model rats.Methods VD rat model was prepared by bilateral common carotid arteries ligation( 2-VO) method.Morris water maze was used to evaluate the learning and remembering ability.The RT-PCR was used to analyse the mRNA expression of Clock,Bmal and Per2 genes on the suprachiasmatic nucleus.Results Tongluo Xingnao Effervescent Tablets can improve learning and memory function of VD rats,and upregulate the expression of Bmal,Clock and Per2 genes in suprachiasmatic nucleus of rats( P〉0.05).Conclusion Tongluo Xingnao Effervescent Tablets can improve the cognitive function of vascular dementia rats,which was relate to the regulation of Bmal,clock and Per2 genes on the suprachiasmatic nucleus.
