摘 要:为获得茄子高效再生体系,本试验以7份茄子材料的无菌苗子叶、下胚轴为外植体,研究了不同激素浓度配比以及不同基因型对茄子离体再生的影响。结果表明:子叶的再生效果优于下胚轴;下胚轴形态学上端再生能力显著高于形态学下端,下胚轴形态学上端出芽率为46%,出芽系数为1~3,下胚轴形态学下端出芽率为0;7份茄子子叶的最优愈伤和不定芽分化培养基均为MS+2 mg/L ZT+0.1 mg/L NAA+30 g/L蔗糖+7 g/L琼脂,出愈率和出芽率均为100%;最优不定芽伸长培养基为MS+30 g/L蔗糖+7 g/L琼脂,伸长率可达93%;7份材料中,徐州长茄和成都墨茄子叶的再生频率较高,分别为0.93和0.73。本研究获得了较高的茄子再生频率,成苗迅速,为利用组织培养技术进行茄子种质创新提供了科学依据。In order to obtain the high-efficiency regeneration system of eggplant, the aseptic seedlings cotyledons and hypocotyls of seven eggplant materials were used as explants to study the effects of different hormone concentrations and different genotypes on the in vitro regeneration of eggplant. The results showed that the regeneration effect of cotyledons was better than that of the hypocotyls. The regeneration capacity of morphological upper end of hypocotyls was significantly higher than that of the morphological lower end. The budding rate of morphological upper end of hypocotyls was 46% with the budding coefficient of 1~3 and 0 of the budding rate of morphological lower end of hypocotyls. The optimum medium for callus induction and adventitious buds differentiation of7 eggplants cotyledons was MS+2 mg/L ZT+0.1 mg/L NAA+30 g/L sucrose+7 g/L agar, and the callus induction rate and budding rate were both 100%. The optimal adventitious buds elongation medium was MS+30 g/L sucrose+7 g/L agar, and the buds elongation rate could be 93%. Among the seven eggplant materials, Xuzhouchangqie and Chengdumoqie had the higher regeneration frequency of cotyledons which was 0.93 and 0.73, respectively. In this study, higher frequency of eggplant regeneration was obtained and the seedling was rapid which would lay an important foundation for the innovation of eggplant germplasm by tissue culture technique.
