摘  要:昆虫细胞色素P450是重要的解毒酶,参与杀虫剂代谢。利用转基因果蝇技术探究舞毒蛾CYP6B53基因对溴氰菊酯和百树菊酯的胁迫响应:通过药膜法以低剂量(0.77 mg/L)溴氰菊酯和百树菊酯胁迫转CYP6B53基因果蝇(命名为att P40>CYP6B53)及非转基因对照果蝇(命名为att P40),分别测定了细胞色素P450酶活性和CYP6B53基因表达量对2种杀虫剂的胁迫响应。结果表明:低剂量拟除虫菊酯胁迫下,转CYP6B53基因果蝇体内细胞色素P450活性表现出不同的时间效应,以24 h时表现为例,溴氰菊酯胁迫24 h显著抑制P450活性,而百树菊酯胁迫则显著诱导P450活性;转基因果蝇体内CYP6B53 m RNA表达水平对2种杀虫剂24 h胁迫响应呈现出相似的结果。综上,CYP6B53基因调控P450活性参与溴氰菊酯和百树菊酯胁迫响应,但胁迫响应存在显著的时间效应。Insect cytochrome P450s are important detoxifying enzymes involved in detoxifying insec- ticides. In this study, we used transformant Drosophila technology to explore the stress response of the CYP6B53 gene of Lymantria dispar to deltamethrin and cyfluthrin stresses. The transformant Dro- sophila expressing CYP6B53 (namely attP40 〉 CYP6B53 ) and the non-transformant Drosophila ( namely attP40) were treated with 0.77 mg/L of dehamethrin and cyfluthrin by the method of resi- due. The cytochrome P450 activities and the CYP6B53 gene expression were determined to stress re- sponse of two kinds of insecticides, respectively. The cytochrome P450 activities of the transformant Drosophila expressing CYP6B53 showed different time effects under the low dose of pyrethroids stres- ses. At 24 h time point, the cytochrome P450 activity was significantly inhibited under deltamethrin stress but significantly induced under cyfluthrin stress. The mRNA expression of CYP6B53 gene in the transformant Drosophila expressing CYP6B53 showed similar effects under two pyrethroids stres- ses. These results indicated that the CYP6B53 gene could regulate the cytochrome P450 activity in response to the stresses of pyrethroids, but the response to the stresses had a significant time effect
