摘 要:目的:观察抗纤灵对5/6肾切除小鼠慢性肾功能衰竭心脏病变NO、ADMA、NT-proBNP的影响,从氧化应激的角度探讨抗纤灵治疗慢性肾功能衰竭心脏病变的作用机制。方法:按Platt法建立慢性肾衰小鼠模型,造模成功后,随机将手术组小鼠分为模型组、缬沙坦组、抗纤灵组。抗纤灵组予抗纤灵方煎剂0.5 m L灌胃,药物浓度为20 g/kg,每日1次;缬沙坦组予缬沙坦0.5 m L灌胃,药物浓度为20 mg/kg,每日1次;模型组和假手术组均予等量蒸馏水灌胃,连续灌胃22周。22周后取材,测量各组小鼠的心脏质量指数,生化法检测小鼠血清NO水平,Elisa法检测ADMA、NT-proBNP水平。结果:与假手术组比较,模型组的心脏质量指数、ADMA、NT-proBNP显著增高(P〈0.01),NO明显降低(P〈0.01)。与模型组比较,抗纤灵组的心脏质量指数有改善(P〈0.05),NO、ADMA、NT-proBNP有显著改善(P〈0.01)。缬沙坦组的心脏质量指数、NO、ADMA、NT-proBNP明显改善(P〈0.01)。抗纤灵组与缬沙坦组组间比较,缬沙坦在改善NT-proBNP和心脏质量指数方面优于抗纤灵组(P〈0.05),抗纤灵组在降低ADMA、升高NO方面优于缬沙坦(P〈0.05)。结论:抗纤灵能通过调节氧化应激水平改善慢性肾功能衰竭引起的心脏病变。Objective:To determine the effect of Kangxianling Decoction(KXLD)on the oxidative stress in the hearts of 5/6 nephrectomized mice model and discuss its mechanism. Methods:C57 BL/6 male mice were performed 5/6 nephrectomization to create chronic renal failure. After the models were established,mice were randomly divided into model group,KXLD group and Valsartan group. KXLD and Valsartan group were respectively administered with 20 g/kg,and Valsartan 20 mg/kg,0.5 m L/d. Model group and sham-operated group were given 0.5 m L water. The mice were sacrificed after 22 weeks of treatment,cardiac weight indices were detected and NO,ADMA,NT-proBNP were examed. Results:Compared with sham group,cardiac weight indices,ADMA,NT-proBNP were significantly increased(P〈0.01)and NO was significantly decreased(P〈0.01)in the model group. Compared with model group,cardiac weight indices were improved(P〈0.05),and NO,ADMA,NT-proBNP were attenuated significantly(P〈0.01)in the KXLD group. Compared with model group,NO,ADMA,NTproBNP and cardiac weight indices were all attenuated significantly(P〈0.01)in the Valsartan group. Compared between Valsartan and KXLD groups,Valsartan was superior to KXLD in cardiac weight indices(P〈0.05)and NT-proBNP,while KXLD was superior to Valsartan in NO,ADMA(P〈0.05). Conclusion:Kangxianling Decoction is able to protect heart function induced by chronic renal failure via adjusting oxidative stress.
