摘 要:基质辅助激光解吸电离飞行时间质谱(matrix-assisted laser desorption ionization time-of-flight mass spectrometry,MALDI-TOF MS)技术是临床微生物诊断中一项革命性创新技术,由于其快速、特异、操作简便等优势,现已迅速被临床实验室接受和认可.然而,MALDI-TOF MS技术在临床应用中仍然存在诸多的挑战,如样品制备还未做到最优化;鉴定技术存在局限性;数据库质量的提高以及微生物种类的添加还在讨论中;抗菌药物敏感试验领域的应用还存在争议;非常见鉴定结果如何转化到临床治疗仍在探索中等.但是MALDI-TOF MS在临床微生物诊断应用中还有一些需改进之处.MALDI-TOF MS is a revolutionary and innovative technologyin clinical microbiological diagnosis. Due to its advantages of speed, specificity and ease of operation, MALDI-TOF MS has been rapidly accepted and approved by clinical laboratory. However, many challenges appeared when applied in clinical application. For example, sample preparation needs to be further optimized. Identification techniques have limitations. The quality improvement of database and the diversity of microbial species are still under discussion. The application of antimicrobial susceptibility testing is still controversial. The way ofrare identification resuhstransmitting into clinical treatment is still being explored. But the application of MALDI-TOF MS in clinical microbiological diagnosis still needs to be improved.
